Effect of Esketamine on Myocardial Injury after Ischemia Reperfusion of Hindlimb in Rats
Objective:To observe the effects of esketamine on myocardial injury(MI)induced by hindlimb ischemia reperfusion(LIR)in rats,and explore the effects of different administration times of esketamine on myocardial injury.Methods:Thirty SD rats were randomly divided into sham surgery group(Sham group),ischemia reperfusion group(IR group),sham surgery+esketamine group(Sham+Esk group),pre-ischemic esketamine+ischemia-reperfusion group(IR+Esk1 group)and pre-reperfusion esketamine+ischemia-reperfusion group(IR+Esk2 group),with 6 rats in each group.Hindlimb ischemia reperfusion model were prepared in other groups except sham group(ischemia 3 h,reperfusion 2 h).After drug treatment,the contents of troponin I(cTnI),tumor necrosis factorα(TNF-α),interleukin-6(IL-6),and norepinephrine(NE)in serum of each group were measured by enzyme-linked immunosorbent assay(ELISA).Results:Compared with Sham group,the contents of serum cTnI,TNF-α,IL-6,and NE contents increased in IR group,while cTnI,TNF-α and IL-6 contents decreased and NE contents increased in Sham+Esk group(P<0.05).Compared with IR group,the contents of serum cTnI,TNF-α and IL-6 contents in IR+Esk1 group and IR+Esk2 group decreased,serum NE content in IR+Esk1 group decreased,and NE in IR+Esk2 group increased,with statistical significance(P<0.05).Conclusion:Esketamine may show a protective effect on myocardial injury induced by hindlimb ischemia reperfusion in rats by inhibiting inflammatory response.
