The Treatment of Autism Neurodevelopmental Disorders with"Changqiang"and"Taixi"Acupoints Electroacupuncture
Objective:To explore the targeted regulatory mechanism of"Changqiang"combined with"Taixi"acupoints electroacupuncture in the treatment of autistic neurodevelopmental disorders in offspring mice.Methods:Ten SD rats were divided into model group and non-model group,with 5 groups in each group.The rats in the model group were given 600 mg/kg sodium valproate by gavage to establish models,and the rats in the non-model group were given the equal volume of normal saline by gavage.The offspring of the rats in the model group were randomly divided into electroacupuncture acupoint group,electroacupuncture non-meridian non-acupoint group and model group with 12 rats,and 12 offspring rats from the non-model group were randomly selected as the control group.The rats in the electroacupuncture acupoint group started electroacupuncture at Changqiang and Taixi acupoints at the 24th day after birth.The electroacupuncture non-meridian non-acupoint group avoided meridian and acupoint acupuncture.Open field experiment and morris water maze test were performed before and after intervention.The hippocampus was collected by decapitation,and the relative expression level of γ-aminobutyric acid type A receptor(GABAAR)mRNA in the hippocampus of the offspring were determined by real-time quantitative PCR(RT-PCR).The relative expression levels of glycogen synthase kinase-3β(GSK-3β),β-catenin and microalbumin(PV)were measured by Western Blot.Results:The open field experiment results showed that after intervention,the horizontal and vertical scores of electroacupuncture acupoint group were higher than those of model group and electroacupuncture non-meridian non-acupoint group,but lower than those of the control group(P<0.05).Morris water maze test results showed that after intervention,compared with the model group and the electroacupuncture non-meridian non-acupoint group,the escape latency of the electroacupuncture point group shortened,the crossing times increased,and the platform residence time prolonged(P<0.05).Compared with model group,the relative expression levels of Glu,GABA,GABAAR mRNA,β-catenin and PV in hippocampus of rats in electroacupuncture acupoint group decreased,while the relative expression levels of GSK-3βprotein increased(P<0.05).Conclusion:Acupuncture at Changqiang and Taixi acupoints can effectively improve the spatial learning and memory ability of autistic offspring mice,and target the regulation of autistic neurodevelopmental disorders.The specific mechanism may be related to promoting the excitation-inhibition balance of GABA neural pathway and acting on important regulatory factors in Wnt/β-catenin signaling pathway.
