The Mechanism of Huoxue Jiedu Jiangtang Recipe for Treating Diabetic Cardiomyopathy Based on Network Pharmacology,Molecular Docking and Experimental Study
Objective:The main chemical components and mechanism of Huoxue Jiedu Jiangtang Recipe for treating diabetic cardiomyopathy(DCM)were studied by network pharmacology,molecular docking and animal experiments.Methods:The active ingredients,core targets and signaling pathways of Huoxue Jiedu Jiangtang Recipe for DCM were determined by means of network pharmacological tools.A DCM rat model was established to verify the main targets predicted by network pharmacology and the therapeutic effect.Results:A total of 158 active ingredients of Huoxue Jiedu Jiangtang Recipe were screened,involving 318 DCM targets and 204 related signaling pathways.Network analysis showed that Huoxue Jiedu Jiangtang Recipe could treat DCM by phosphatase gene(PTEN),target protein of rapamycin(mTOR),tumor protein P53(TP53),signal transduction and transcriptional activator 3(STAT3),vascular endothelial cell growth factor(VEGFA),and other key targets.Molecular docking showed that Huoxue Jiedu Jiangtang Recipe showed better binding ability with quercetin,γ-aminobutyric acid,adenosine,luteolin,kaempferol and PTEN,mTOR,TP53,STAT3,VEGFA,and other key targets.The vina scores of luteolin and mTOR were the lowest.Animal experiments showed that Huoxue Jiedu Jiangtang Recipe could alleviate myocardial fibrosis,reduce the levels of C-reactive protein(CRP),tumor necrosis factor-α(TNF-α),interleukin(IL)-6,and inhibit the expression of mTOR protein,so as to play some role for the treatment of DCM.Conclusion:Huoxue Jiedu Jiangtang Recipe could improve DCM by multi-component and multi-target way,which provided valuable research directions for further investigation of its therapeutic effects.
