CircACAP2靶向miR-29a-3p/CCNT2轴对缺氧/复氧诱导的心肌细胞损伤的影响

目的:探讨 CircACAP2靶向 miR-29a-3p/细胞周期蛋白 T2(CCNT2)轴对缺氧/复氧(H/R)诱导的心肌细胞损伤的影响。方法:将 H9c2细胞分为Control组(正常培养)、si-NC组(转染si-NC)、Model组(建立H/R模型)、si-CircACAP2组(转染si-CircACAP2)、si-CircACAP2+inhibitor-NC组(si-CircACAP2和inhibitor-NC共转染)、si-CircACAP2+miR-29a-3p inhibitor组(si-CircACAP2和miR-29a-3p inhibitor共转染);实时荧光定量聚合酶链式反应(qRT-PCR)检测 H9c2 细胞中 CircACAP2、miR-29a-3p的表达水平;四唑盐(MTT)法检测 H9c2 细胞活力;流式细胞仪检测 H9c2 细胞凋亡;单丹磺酰尸胺(MDC)染色法检测 H9c2 细胞自噬;乳酸脱氢酶(LDH)、丙二醛(MDA)、超氧化物歧化酶(SOD)试剂盒检测 H9c2细胞上清中 LDH、MDA、SOD水平,DCFH-DA荧光探针法检测细胞内活性氧(ROS)水平;蛋白质免疫印迹法(Western Blot)检测增殖细胞核抗原(PCNA)、细胞凋亡蛋白[B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl-2相关 X蛋白(Bax)]、自噬相关蛋白(Beclin-1、P62)及 CCNT2 的表达;双荧光素酶报告检测 CircACAP2 对 miR-29a-3p的靶向关系和 miR-29a-3p对 CCNT2的靶向关系。结果:与 Control组比较,Model组 H9c2 细胞中 CircACAP2 表达、凋亡率、自噬囊泡数、LDH释放率、MDA、ROS水平、Bax、Beclin-1、CCNT2 蛋白表达升高,miR-29a-3p表达、OD490 值、SOD活性、PCNA、Bcl-2、P62 蛋白表达降低(P<0。05);与 Model 组比较,si-CircACAP2 组 H9c2 细胞中 CircACAP2 表达、凋亡率、自噬囊泡数、LDH 释放率、MDA、ROS水平、Bax、Beclin-1、CCNT2蛋白表达降低,miR-29a-3p表达、OD490值、SOD活性、PCNA、Bcl-2、P62蛋白表达升高(P<0。05);与si-CircACAP2组比较,si-CircACAP2+miR-29a-3p inhibitor组 H9c2细胞中 CircACAP2表达差异无统计学意义(P>0。05),凋亡率、自噬囊泡数、LDH释放率、MDA、ROS水平、Bax、Beclin-1、CCNT2 蛋白表达升高,miR-29a-3p表达、OD490值、SOD活性、PCNA、Bcl-2、P62蛋白表达降低(P<0。05);CircACAP2 靶向调控 miR-29a-3p 的表达,miR-29a-3p 靶向负调控 CCNT2 的表达。结论:敲低CircACAP2可能通过靶向miR-29a-3p来下调CCNT2表达,进而抑制H/R诱导的H9c2细胞凋亡、自噬能力及氧化应激,促进细胞增殖,发挥保护作用。
Effect of CircACAP2 Targeting miR-29a-3p/CCNT2 Axis on Hypoxia/Reoxygenation Induced Cardiomyocyte Injury
Objective:To investigate the effect of CircACAP2 targeting miR-29a-3p/cyclin T2(CCNT2)axis on hypoxia/reoxygenation(H/R)induced cardiomyocyte injury.Methods:H9c2 cells were divided into Control group(normal culture),si-NC group(transfected with si-NC),Model group(established H/R model),si-CircACAP2 group(transfected with si-CircACAP2),si-CircACAP2+inhibitor-NC group(si-CircACAP2 and inhibitor-NC co-transfection),si-CircACAP2+miR-29a-3p inhibitor group(si-CircACAP2 and miR-29a-3p inhibitor co-transfection).The expression levels of CircACAP2 and miR-29a-3p,cell viability and apoptosis of H9c2 cells were detected.H9c2 autophagy,lactate dehydrogenase(LDH),malondialdehyde(MDA),superoxide dismutase(SOD),reactive oxygen species(ROS)levels and expression of proliferating cell nuclear antigen(PCNA),B-cell lymphoma/leukemia-2(Bcl-2),Bcl-2 associated X protein(Bax),Beclin-1,P62 CCNT2 were dected.The targeting relationship of CircACAP2 to miR-29a-3p and the targeting relationship of miR-29a-3p to CCNT2 were detected.Results:Compared with the Control group,the expression of CircACAP2,apoptosis rate,number of autophagy vesicles,LDH release rate,MDA,ROS levels and Bax,Beclin-1,CCNT2 protein increased in H9c2 cells in Model group,the expression of miR-29a-3p,OD490 value,SOD activity,PCNA,Bcl-2 and P62 protein expression decreased(P<0.05).Compared with Model group,CircACAP2 expression,apoptosis rate,number of autophagy vesicles,LDH release rate,MDA,ROS levels,Bax,Beclin-1 and CCNT2 protein expression in H9c2 cells of si-CircACAP2 group decreased,miR-29a-3p expression,OD490 value,SOD activity,PCNA,Bcl-2,P62 protein expression increased(P<0.05).Compared with si-CircACAP2 group,there was no statistically significant difference in CircACAP2 expression in H9c2 cells of si-CircACAP2+miR-29a-3p inhibitor group(P>0.05),apoptosis rate,number of autophagy vesicles,LDH release rate,MDA,ROS levels and Bax,Beclin-1,CCNT2 protein expression increased,and miR-29a-3p expression,OD490 value,SOD activity,PCNA,Bcl-2,P62 protein expression decreased(P<0.05).CircACAP2 targeted the expression of miR-29a-3p,and miR-29a-3p negatively regulated the expression of CCNT2.Conclusion:Knockdown of CircACAP2 may down-regulate CCNT2 expression by targeting miR-29a-3p,thereby inhibiting H/R-induced apoptosis,autophagy and oxidative stress of H9c2 cells,promoting cell proliferation and playing a protective role.
