Effects of Hengqing Ⅴ Prescription on Epilepsy Mice Induced by Kainic Acid
Objective:To explore effects of Hengqing Ⅴ Prescription on epilepsy mice induced by kainic acid.Methods:Forty male wild-type C57BL/6J mice were randomly divided into normal control group,model group,Hengqing Ⅴ Prescription intervention group,and valproate intervention group,with 10 mice in each group.The mice in intervention group of Hengqing Ⅴ Prescription were administrated with Hengqing Ⅴ Prescription by gavage.The mice in valproate intervention group were administrated with valproate by intraperitoneal injection,and the mice in normal control group and model group were administrated with physiological saline by gavage,once a day for 10 days.After the last administration,except for normal control group,the other groups were injected intraperitoneal with kainic acid to induce seizures.The behavioral changes and spatial working memory ability were observed.After the observation,the levels of malondialdehyde(MDA),superoxide dismutase(SOD),catalase(CAT),and total antioxidant capacity(T-AOC)in the hippocampus of mice in each group were measured.Results:Compared with normal control group,model group mice showed significantly longer duration of epileptic state,shorter latency of epilepsy,higher frequency of seizures above grade 3,and less number of new arm explorations,time of new arm explorations,and exploration distance of new arm explorations(all P<0.01).Compared with model group,the duration of epileptic state in Hengqing Ⅴ Prescription intervention group and the intervention group of valproate significantly shortened,the latency time of epilepsy prolonged,the number of seizures above grade 3 reduced,number of new arm explorations,time of new arm explorations,and exploration distance of new arm explorations all increased(all P<0.01).Compared with valproate intervention group,mice in the Hengqing Ⅴ Prescription intervention group showed a shorter duration of epileptic state,a longer latency of epilepsy,fewer seizures above grade 3,and an increase in the number,time,distance of explorations in new arm explorations(all P<0.05).Compared with normal control group,MDA level in the hippocampal tissue of model group mice increased,while the levels of SOD,CAT,and T-AOC decreased(all P<0.01).Compared with model group,the levels of MDA in the hippocampus of mice in Hengqing Ⅴ Prescription intervention group of and valproate intervention group were significantly reduced,while the levels of SOD,CAT,and T-AOC were significantly increased(all P<0.01).Compared with valproate intervention group,MDA level in the hippocampus tissue of mice in the Hengqing Ⅴ Prescription intervention group decreased,while the levels of SOD,CAT,and T-AOC increased(all P<0.05).Conclusion:Hengqing Ⅴ Prescription can intervene seizures in kainic acid-induced mice,improve seizure symptoms,and improve cognitive function.Its effect is superior to that of valproate.Hengqing Ⅴ Precription may exert its anti-epileptic effect through anti-oxidative stress.
