Effects of Buyang Huanwu Decoction on SIRT1/HMGB1/NF-κB Pathway and Inflammation in Atherosclerotic Mice
Objective:To observe the effects of Buyang Huanwu Decoction on the signaling pathway of silence-regulator-related enzyme 1(SIRT1)/high mobility group protein B1(HMGB1)/nuclear transcription factor-κB(NF-κB) and inflammation in apolipoprotein E gene knockout(ApoE-/-) mice model of atherosclerosis.Methods:After 2 weeks of adaptive feeding,ApoE-/-mice were fed high-fat diet to establish atherosclerosis model.After 8 weeks,mouse aortic roots were isolated and paraffin sections were prepared.The model was successfully replicated by oil red O staining.Forty ApoE-/-mice were randomly divided into model group,Buyang Huanwu Decoction low-dose group,Buyang Huanwu Decoction medium-dose group and Buyang Huanwu Decoction high-dose group,with 10 mice in each group.In addition,10 C57BL/6J mice were set as blank group by continuing the normal feed,and each group of mice were intervened for 4 weeks.The expression levels of serum inflammatory factors[γ interferon(IFN-γ),tumor necrosis factor α(TNF-α),interleukin(IL)-12,IL-1β]were detected by enzyme-linked immunosorbent assay(ELISA).The expressions of SIRT1,HMGB1 and NF-κB protein in mouse aorta were detected by Western Blot.Results:Buyang Huanwu Decoction could increase the expression level of SIRT1 protein in mice aortic plaques and reduce the expression level of HMGB1 and NF-κB protein,reduce the expression level of IFN-γ,TNF-α,IL-12 and IL-1β in mice serum. Conclusion:Buyang Huanwu Decoction could increase the expression of SIRT1 protein in mouse aortic plaque,decrease the expression of HMGB1,NF-κB protein,and the expression of serum IFN-γ,TNF-α,IL-12,and IL-1β.
