采用随机扩增多态性DNA(Random Amplified Polymorphism DNA,RAPD)技术,运用集群分离分析法(Bulked Segregant Analysis,BSA)研究与罗汉果性别连锁的分子标记.从90条随机引物的扩增产物中筛选出18条可能与性别连锁的RAPD标记.利用雌雄个体对这18条RAPD标记进行验证,发现引物S1431所扩增的约400bp谱带在8个雌性单株中都不出现,而在8个雄性单株中都出现,表明这是一条与雄性连锁的RAPD标记.
Study on RAPD Marker Linked to Sex in Siraitia grosvenorii
RAPD (Random Amolified Polymorphism DNA) was employed to detect molecular markers linked to sex in S. grosve-norii by BSA (Bulked Segregant Analysis). 18 RAPD markers linked to sex were selected in BSA by screening 90 primers. Only the marker amplified by S1431 was present in all 8 male individuals tested while absent from all 8 female individuals tested. It showed that S1431 was a male RAPD marker linked to sex.
