目的:研究活血化瘀中药鸡血藤对小鼠黑色素瘤高转移细胞B16-BL6生长、黏附、侵袭、运动、转移能力的影响,并初步探讨其作用机制.方法:采用MTT法测定鸡血藤对B16-BL6细胞增殖能力的影响,采用细胞黏附实验、重组基底膜侵袭实验、趋化性运动能力实验以及小鼠黑色素瘤自发性转移模型,观察鸡血藤对B16-BL6细胞生长、黏附、侵袭、运动及转移能力的影响.结果:鸡血藤水提液在最高无毒浓度下,对B16-BL6细胞与基底膜成分LN的黏附及侵袭基底膜能力具有显著的抑制作用,但对穿越基底膜的运动能力无显著作用;高剂量鸡血藤能显著减少小鼠肺转移结节数目与直径,减轻肺转移程度.结论:鸡血藤提取液能抑制B16-BL6的转移,该作用可能与其抑制B16-BL6细胞与细胞外基质的黏附以及对基底膜的侵袭能力有关.
Effect of Spatholobus suberctus on Adhesion, Invasion, Migration and Metastasis of Melanoma Cells
Objective:To study the effect of Spatholobus suberctus,a kind of Chinese Traditional Medicine which can dissolve the stasis by activating the blood circulation, on invasion, adhesion, migration and metastasis of B16-BL6 metastatic mouse melanoma cells and its mechanism. Methods:The proliferation,adhesion,invasion and migration capacity of B16-BL6 metastatic cells was evaluated by MTT assay,adhesion assay and reconstituted basement membrane invasion and migration assay in vitro respectively. Mouse spontaneous motilily melanoma model was used to study the effect of Spatholobus suberctus on metastasis in vivo. Results:At the highest innoxious concentration,the extracts of Spatholobus suberctus inhibited the adhesion and invasion capacity of B16-BL6 metastatic cells significantly. In the mouse spontaneous melanoma model,the lung metastatic nodes number and its volume were significantly decreased after continuously treated with the extracts of Spatholobus suberctu. Conclusion:The extracts of Spatholobus suberctu can inhibit the metastasis of of B16-BL6 metastatic mouse melanoma cells and its mechanism may be inhibiting the capability of B16-BL6 cells in adhering to the ECM and invading the basement membrane.
