Extraction and Analysis of the Essential Oil in Pogostemon cablin by Enzymatic Hydrolysis and Inhibitory Activity Against Hela Cell Proliferation
Objective:To optimize the extraction method of essential oil in Pogostemon cablin and analyze its inhibitory activity a-gainst Hela cell proliferation . Methods:The Pogostemon cablin was treated by hemicellulase before steam distillation. The enzyme dosage, treatment time,treatment temperature,pH were optimized through orthogonal experimental design. The components of essential oil were identified by gas chromatography-mass spectrometry (CC-MS). Inhibitory activity of patchouli oil against Hela cell proliferation was determined by MIT method Results:The optimum extraction process was as follows; pH 4.5 ,temperature 45 ℃, the ratio of hemicellulase to Pogostemon cablin was 1% and enzymatic hydrolysis for 1.0 hour. Extraction ratio of the patchouli oil in steam distillation and hemicellulase extraction method was 2. 222 0 mg/g,3. 136 0 mg/g respectively. Patchouli oil could inhibit Hela cell proliferation. IC50 of the patchouli oil in steam distillation and hemicellulase extraction method was 12.2 ±0.46 μg/mL and 0. 36 ±0.03 μg/mL respectively. Conclusion:In comparison with steam distillation method,extraction ratios of essential oil and the inhibitory activity against Hela cell proliferation can be increased by the hemicellulase extraction method.
