Effect of Essential Oil from Schizonepeta tenuifolia on the Mechanism of NLRP3 Inflammasome Activation in THP-1 Cells
Objective:To study the mechanism of the anti-inflammatory effect of essential oil from Schizonepeta tenuifolia on NL-RP3 inflammasome activation in THP-1 cells.Methods:Toxicity of the essential oil from Schizonepeta tenuifolia was detected by MTS method.THP-1 cells were differentiated into macrophages with PMA,then treated with LPS and activators (ATP,Nigericin,CPPD and CaC12) to activate NLRP3 inflammasome.The effect of essential oil from Schizonepeta tenuifolia on NLRP3 inflammasome were observed.The level of IL-1β and IL-18 in supernatant were measured by ELISA.RT-PCR was performed to observe the mRNA expression of NLRP3,Caspase-1,IL-1β,IL-1 α and IL-6.Results:The maximum non-toxic concentration of esential oil from Schizonepeta tenuifolia was 0.4 mg/mL.0.2 mg/mL essential oil from Schizonepeta tenuifolia could significantly reduce the level of IL-1β which induced by ATP,Nigericin,CPPD or CaCl2 (P < 0.05 or P < 0.01),and could inhibit the mRNA high expression of NLRP3,Caspase-1,IL-1β,IL-1a and IL-6 in THP-1 cells which induced by ATP or Nigericin(P < 0.05 or P < 0.01).Conclusion:Essential oil of Schizonepeta tenuifolia can reduce the high level of IL-1β which induced by ATP,Nigericin,CPPD or CaC12,and inhibit the mRNA high expression of NLRP3,Caspase-1,IL-1β,IL-1α and IL-6 which induced by ATP or Nigericin.The anti inflammatory mechanism of the essential oils from Schizonepeta tenuifolia is related to inhibiting the activating of NLRP3 inflammasome.
