目的:研究玉竹地上部位的化学成分及其对α-葡萄糖苷酶的抑制活性.方法:采用大孔吸附树脂HP-20、Sephadex LH-20、MCI gel CHP 20P、Chromatorex C18、半制备型高效液相等色谱法对玉竹地上部位75%甲醇提取物进行分离纯化,通过1 H-NMR、13C-NMR等波谱技术对化合物进行结构鉴定,并采用α-葡萄糖苷酶活性筛选模型对部分化合物进行活性筛选.结果:从玉竹地上部位中分离得到16个化合物,分别鉴定为:对羟基苯甲酸(1)、芹菜素-6-C-葡萄糖苷(2)、异金雀花素(3)、异槲皮苷(4)、肥皂草苷(5)、山柰酚-3-O-β-芸香苷(6)、芦丁(7)、异鼠李素-3-O-芸香苷(8)、牡荆素-2″-O-葡萄糖苷(9)、牡荆素-4″-O-葡萄糖苷(10)、牡荆素-2″-O-β-D-(6′″-乙酰基)-葡萄糖苷(11)、异牡荆素-2″-O-β-D-葡萄糖苷(12)、芹菜素-6-C-阿拉伯糖-葡萄糖苷(13)、槲皮素-7-O-[α-L-鼠李糖-(1→6)-β-D-半乳糖苷](14)、金圣草黄素-7-O-槐糖苷(15)、金圣草黄素-7-O-[β-D-芹菜糖-(1→6)]-β-D-葡萄糖苷(16),并筛选了部分化合物的α-葡萄糖苷酶抑制活性.结论:其中,化合物2~6、10~16为首次从该植物中分离得到.化合物2具有显著的α-葡萄糖苷酶抑制活性.
Study on the Chemical Constituents from the Aerial Parts of Polygonatum odoratum and α-Glucosidase Inhibitory Activity
Objective:To study the chemical constituents from the aerial parts of Polygonatum odoratum and the inhibition on α-glucosidase activity.Methods:The 75%methanol extract from the aerial parts of Polygonatum odoratum was isolated and purified by va-rious chromatographic techniques,such as macroporous resin HP-20,Sephadex LH-20,MCI gel CHP 20P,Chromatorex C18,semi-pre-parative HPLC and others.The structures were identified on the basis of 1H-NMR,13C-NMR.In addition,the α-glucosidase inhibitory activity of the compounds were evaluated by pharmacological methods simultaneously.Results:Sixteen compounds were isolated from the aerial parts of Polygonatum odoratum and identified as p-hydroxybenzoic acid(1),6-C-glucosylapigenin(2),isoscoparine(3),isoquer-citrin(4),saponarin(5),kaempferol-3-O-β-rutinoside(6),rutin(7),isorhamnetin-3-O-rutinoside(8),2″-O-glucosylvitexin(9),4″-O-glucosylvitexin(10),vitexin-2″-O-β-D-(6′″-acetyl)-glucoside(11),isovitexin-2″-O-β-D-glucoside(12),apigenin-6-C-arabinosylglu-coside(13),quercetin-7-O-[α-L-rhamnopyranosyl(1→6)-β-D-galactopyranoside](14),chrysoeriol-7-O-sophoroside(15),7-O-[β-D-glicopiranosil-(1→6)-β-D-apiofuranosil]crisoeriol(16),respectively.Some of the compounds were also screened for α-glucosidase in-hibitory activity.Conclusion:Among them,compounds 2 to 6,10 to 16 are isolated from Polygonatum odoratum for the first time,com-pound 2 shows significant inhibitory effect on α-glucosidase.
