摘要
目的:研究五福花科荚蒾属植物台东荚蒾Viburnum taitoense Hayata的化学成分及其抗炎活性.方法:通过柱色谱和半制备液相色谱等方法对台东荚蒾的乙酸乙酯部位进行分离纯化,利用现代波谱技术鉴定化合物结构,通过检测脂多糖(LPS)诱导小鼠巨噬细胞(RAW264.7)一氧化氮(NO)的释放,评价化合物的抗炎活性.结果:从台东荚蒾中分离鉴定了 14 个化合物,分别为:3β-acetoxy-17β-hydroxy-28-norolean-12-ene(1)、3-O-乙酰齐墩果醛(2)、3β-hydroxy-3-deoxymoronic acid(3)、6β-hydroxy-3-oxo-11,13(18)-oleanadien-28-oic acid(4)、3-O-乙酰齐墩果酸(5)、3β-羟基-18-烯-齐墩果烷(6)、科罗索酸(7)、3β,6β-二羟基齐墩果-11,13(18)-二烯-28-酸(8)、熊果醇(9)、3β-羟基-齐墩果烷-11,13(18)-二烯-28-酸(10)、槲皮素-3-O-α-L-鼠李糖苷(11)、山柰酚-3-O-α-L-鼠李糖苷(12)、原儿茶酸(13)、香草酸(14).结论:其中,化合物 1~3 为首次从荚蒾属植物中分离得到,化合物 4~6、12~14 为首次从该植物中分离得到.化合物 7 表现出较强的抗炎活性,其IC50 值为(6.12±0.32)μmol/L,明显优于阳性药地塞米松,化合物 4、5、8~10 表现出一定的抗炎活性.
Abstract
Objective:To study the chemical constituents and anti-inflammatory activity of Viburnum taitoense.Methods:The chemi-cal constituents from the ethyl acetate extract of Viburnum taitoense were isolated and purified by various column chromatography and semi-preparative liquid methods.The stuctures were identified by modern spectral techniques.The anti-inflammatory activities were e-valuated by against NO production form LPS stimulated RAW264.7 cell.Results:Fourteen compounds were isolated from Viburnum tai-toense and identified as 3β-acetoxy-17β-hydroxy-28-norolean-12-ene(1),3-O-acetyl oleanolic aldehyde(2),3β-hydroxy-3-deoxymoronic acid(3),6β-hydroxy-3-oxo-11,13(18)-oleanadien-28-oic acid(4),3-O-acetyloleanolic acid(5),3β-hydroxy-olean-18-ene-germanicol(6),corosolic acid(7),3β,6β-dihydroxy oleana-11,13(18)-dien-28-acid(8),uvaol(9),3β-hydroxy-oleana-11,13(18)-dien-28-oic acid(10),quercetin-3-O-α-L-rhamnoside(11),kaempferol-3-O-α-L-rhamnoside(12),protocatechuic aicd(13),vanillic acid(14),re-spectively.Conclusion:Among them,compounds 1 to 3 are isolated from Viburnum genus for the first time,compounds 4 to 6,12 to 14 are isolated from Viburnum taitoense for the first time.Compound 7 shows strong anti-inflammatory activity with IC50 value of(6.12±0.32)μmol/L,significantly better than the positive drug dexamethasone.Compounds 4,5,8 to 10 show moderate anti-inflammatory ac-tivities.
