基于网络药理学探讨蠲哮汤治疗哮喘的作用机制

目的:运用网络药理学方法探讨蠲哮汤对哮喘的防治作用及可能的分子机制.方法:利用中药系统药理数据库和分析平台(TCMSP)筛选蠲哮汤药物活性成分及作用的靶蛋白,通过PubChem 和Swiss Target Prediction数据库收集活性成分作用靶点的基因名,从GeneCards和OMIM数据库中获得哮喘疾病相关靶点,利用Venny 2.1.0 获得的二者交集靶点即为蠲哮汤治疗哮喘的潜在作用靶点.利用Cytoscape 3.9.0 软件构建"中药-活性成分-作用靶点"网络图,采用STRING数据库构建蠲哮汤治疗哮喘潜在作用靶点的蛋白质-蛋白质相互作用网络,并利用Cytoscape 3.9.0 软件对其进行拓扑学分析,筛选出核心靶点.进一步利用DAVID数据库对筛选得到的靶点进行基因本体论(GO)和京都基因与基因组百科全书(KEGG)富集分析,进一步筛选重要化合物与疾病靶点,进行分子对接预测.结果:共得到蠲哮汤 60 个潜在活性成分,预测到 110 个作用靶点.收集哮喘靶点 1432 个,取交集得到蠲哮汤治疗哮喘的作用靶点 36 个,对应活性成分 23 个,其中以槲皮素、山柰酚、芦丁和柚皮素为主.拓扑分析得到 36 个节点、144 条作用关系,丝氨酸/苏氨酸蛋白激酶 1(Akt Serine/Threonine Kinase 1,AKT1)、表皮生长因子受体(Epidermal Growth Factor Receptor,EGFR)、过氧化物酶体增殖物激活受体-γ(Peroxisome Proliferator Activated Receptor Gamma,PPARG)、SRC原癌基因(SRC Proto-Oncogene,SRC)等可能为关键靶点.GO富集分析得到 160 个条目,其中生物过程条目 115 个、分子功能条目 31 个、细胞组分条目 14 个;KEGG富集到通路 51 条,主要涉及癌症中的蛋白多糖、雌激素信号通路、磷脂酰肌醇 3 激酶(Phosphatidylinositol 3-kinase,PI3K)-蛋白激酶B(Akt)信号通路、血管内皮生长因子(Vascular Endothelial Growth Factor,VEGF)信号通路等.分子对接结果显示,关键成分与核心靶点蛋白有良好的结合活性,证明了网络药理学预测结果的可靠性.结论:本研究首次预测了蠲哮汤通过中药多成分、多靶点、多通路发挥治疗哮喘的作用.经蛋白质-蛋白质相互作用网络分析、GO分析、KEGG富集分析和分子对接验证发现,蠲哮汤可能是通过槲皮素、山柰酚、柚皮素、β-谷甾醇、橙皮苷、芦丁等活性成分,作用于AKT1、EGFR、PPARG、SRC等靶点,阻滞癌症中的蛋白多糖通路、雌激素信号通路、PI3K-Akt和VEGF信号通路发挥诱导平滑肌细胞增殖、细胞生长和分化、黏液分泌等生理过程,抑制细胞外基质沉积、新生血管生成和VEGF,以减轻哮喘气道炎症和气道重塑症状,也为进一步研究蠲哮汤治疗哮喘气道重塑作用机制提供了理论基础.
Discussion on the mechanism of the Juanxiao decoction in treating asthma based on network pharmacology
Objective:Exploring prevention and treatment effects of the Juanxiao decoction(蠲哮汤)on asthma and underlying molecular mechanisms by network pharmacology.Methods:The effective chemical ingredients and relevant targets of drugs in the Juanxiao decoction were retrieved in the TCMSP.The Swiss Target Prediction database and PubChem database were used to collect the name of targets of potential active ingredients.The targets related to asthma were obtained from GeneCards and OMIM,and common targets were obtained by intersection analysis from Venny 2.1.0.The single medicine-active components-targets network was constructed by the software of Cytoscape 3.9.0.The PPI network was constructed by STRING data platform,and the topological analysis was performed by Cytoscape 3.9.0 to obtain the core targets.Furthermore,DAVID database was used to enrich and analyze the GO and KEGG pathways of the selected targets.Molecular docking validation of active ingredients and core targets was performed by molecular docking technology.Results:A total of 60 potential active ingredients of the Juanxiao decoction were obtained by screening TCMSP database.A total of 110 targets of potential active ingredients and 1432 targets related to asthma were screened out.And 36 targets of the Juanxiao decoction for treating asthma were obtained by intersecting which corresponds to 23 active ingredients of the Juanxiao decoction,mainly including quercetin,kaempferol,rutin and naringenin.Topology analysis of the common targets yielded 36 nodes and 144 edges,and AKT1,EGFR,SRC,PPARG could be the key targets.Finally,160 GO terms of the Juanxiao decoction on asthma(including 115 biological processes,31 molecular functions and 14 cell components)were gained.And 51 pathways were identified by KEGG analysis,indicating that the Juanxiao decoction mainly played a role in treating asthma by regulating proteoglycans in cancer,estrogen signaling pathway,PI3K-Akt signaling pathway and VEGF signaling pathway.The results of macromolecular docking showed that key components had good binding activity to core protein targets,which proved the reliability of network pharmacology prediction results.Conclusion:The study predicts for the first time that the mechanism of the Juanxiao decoction may be multi-component,multi-target,multi-pathway in treating asthma.PPI Network analysis,GO analysis,KEGG enrichment analysis and macromolecular docking verification found that the Juanxiao decoction may act on AKT1,EGFR,PPARG,SRC through quercetin,kaempferol,β-sitosterol,naringenin,hesperidin,rutin,and so on to block the proteoglycan pathway in cancer,estrogen pathway,PI3K-Akt and EGFR pathway to induce the smooth muscle cell proliferation,cell growth and differentiation,mucus secretion and other physiological processes,thereby inhibiting extracellular matrix deposition,angiogenesis and VEGF,so as to reduce airway inflammation and airway remodeling in asthma.It also provides a theoretical basis for further research on the mechanism of the Juanxiao decoction in the treatment of asthma airway remodeling.
