A comparative study on treating hypertrophic scar in rabbit model with quintuple scar cream and silicone cream
Objective:To explore the comparative study of quintuple scar cream and silicone cream in the treatment of hypertrophic scar.Methods:A total of 27 healthy cleaning New Zealand large ear white rabbits were selected,and a hypertrophic scar model was established on the ventral side according to the long axis to avoid large blood vessels.Six circular wounds were established in each rabbit ear,with a total of 324 wounds.The random number table method was used to divided them into the control group,the quintuple scar cream group(after the verification of the concentration of 25%,50%and 75%quintuple scar cream in the early stage,quintuple scar cream with the concentration of 50%was used finally),and the silicone cream group,with 9 rabbits in each group.After the operation,disinfection was performed.After 28 days,when the wound was completely epithelialized,the same amount of cream was evenly applied to the scar surface,twice a day,for 28 days of treatment.The results of scar hyperplasia index,fibroblast number density,collagen fibrotic surface density,type Ⅰ and type Ⅲ collagen protein immunohistochemistry and Western blotting were compared among the three groups.Results:The scar hyperplasia index,fibroblast number density and collagen fiber surface density in the silicone cream group were significantly lower than the quintuple scar cream group and the control group,and the above indicators in the quintuple scar cream group was lower than the control group.The differences among the three groups were significant(P<0.05).Immunohistochemical results and Western blotting detection showed that the relative expressions of type Ⅰ collagen and type Ⅲ collagen in the silicone cream group were significantly lower than those in the quintuple scar cream group and the control group,and those in the quintuple scar cream group was lower than the control group,with statistical significant differences among the three groups(P<0.05).Conclusion:Both quintuple scar cream and silicone cream show remarkable effects on hypertrophic scar,both of which can significantly improve the scar tissue color,reduce the scar thickness and soften it,reduce the number of fibroblasts,collagen fiber aggregation and the expressions of type Ⅰ collagen and type Ⅲ collagen of hypertrophic scar,prevent and treat scar,and then promote the recovery,but silicone cream shows more obvious advantages.
