基于网络药理学探讨溃愈灵膏治疗糖尿病足的作用机制

目的:基于网络药理学探讨溃愈灵膏治疗糖尿病足的可能作用机制.方法:基于中药系统药理学数据库与分析平台(TCMSP)、本草组鉴(HERB)数据库获取溃愈灵膏药物活性成分和作用靶点,通过人类基因数据库(GeneCards)和人类孟德尔遗传数据在线系统(OMIM)数据库收集糖尿病足的疾病相关靶点,将药物活性成分作用靶点与疾病相关靶点相映射,获得的交集靶点即为溃愈灵膏治疗糖尿病足的潜在作用靶点.使用CytoScape 3.8.0软件进行"活性成分-交集靶点"网络构建,通过STRING数据库构建蛋白质-蛋白质相互作用网络,分析后筛选出核心靶点,利用R4.2.1软件对交集靶点进行基因本体论(GO)及京都基因与基因组百科全书(KEGG)通路的富集分析.结果:筛选得到99个活性成分及364个潜在作用靶点;获得槲皮素、山达海松酸、异海松酸、积雪草酸、山柰酚等核心活性成分;丝裂原活化蛋白激酶1(Mitogen-ActivatedProtein Kinases 1,MAPK1)、表皮生长因子受体(Epidermal Growth Factor Receptor,EGFR)、丝氦酸/苏氦酸蛋白激酶 1(Akt Serine/Threonine Kinase 1,AKT1)、雌激素受体1(Estrogen Receptor 1,ESR1)等核心靶点;共筛选得到信号通路170条,获得磷脂酰肌醇3激酶(Phosphatidylinositol 3-kinases,PI3K)-蛋白激酶B(Akt)信号通路、MAPK信号通路、糖尿病并发症中的晚期糖基化终末产物(Advanced Glycation End Products,AGE)-晚期糖基化终末产物受体(Advanced Glycation End Product Receptor,RAGE)信号通路等关键通路.结论:溃愈灵膏可能通过槲皮素、山达海松酸、异海松酸等活性成分,作用于MAPK1、AKT1、EGFR、ESR1等潜在核心靶点,调控PI3K-Akt信号通路、MAPK信号通路、AGE-RAGE信号通路等关键通路治疗糖尿病足.
Discussion on the mechanism of Kuiyuling ointment in the treatment of diabetic foot based on network pharmacology
Objective:To explore the possible mechanism of Kuiyuling ointment(清愈灵膏)in the treatment of diabetic foot based on network pharmacology.Methods:Based on the TCMSP and HERB databases,the active ingredients and targets of Kuiyuling ointment were obtained,and the disease-related targets of diabetic foot were collected through GeneCards and OMIM databases.Point-to-point mapping,the obtained intersection target is the potential target of Kuiyuling ointment in the treatment of diabetic foot.CytoScape 3.8.0 software was used to construct an active ingredient-intersection target network,and a PPI network was constructed through the STRING database.Targets were subjected to GO function and KEGG pathway enrichment analysis.Results:A total of 99 active ingredients and 364 potential targets were screened,and core active ingredients such as quercetin,sandapic acid,isopimaric acid,asiatic acid and kaempferol were obtained,core active ingredients such as MAPK1,AKTl,EGFR,ESR1 were obtained.A total of 170 signaling pathways were screened,and key pathways such as PI3K-Akt signaling pathway,MAPK signaling pathway,and AGE-RAGE signaling pathway were obtained.Conclusion:Kuiyuling ointment may act on potential core targets such as MAPK1,AKT1,EGFR,ESR1 through active ingredients such as quercetin,sanda-pimamate,and isopimaric acid,and regulate key pathways such as PI3K-Akt signaling pathway,MAPK signaling pathway,AGE-RAGE signaling pathway in the treatment of diabetic foot.
