A study on the clinical significance of the peripheral blood T cell subsets percentage and the expression of IL-17 and FOXP3 in skin lesions in patients with chronic actinic dermatitis
Objective:To detect the percentage of Th17,Treg,Th1 and Th2 cells in peripheral blood of patients with chronic actinic dermatitis(CAD)and the expression of IL-17 and FOXP3 in skin lesions,and to explore the change trend and related clinical significance.Methods:Peripheral blood T cell subsets were detected.A total of 20 patients who met CAD diagnostic criteria in dermatology department of the First Affiliated Hospital of Dali University were selected as the experimental 1 group.The control 1 group was selected from 20 healthy subjects who met the study criteria in the physical examination department.The percentage of Th1,Th2,Th 17 and Treg cells in peripheral blood were detected by flow cytometry,and the ratio of Th1/Th2 and Th17/Treg were calculated.Corresponding statistical methods were used for statistical analysis.IL-17 and FOXP3 in skin lesions were detected.A total of 20 patients who met CAD diagnostic criteria in dermatology department of the First Affiliated Hospital of Dali University were selected as the experimental 2 group.Twenty cases of normal skin tissue around the nevus tissue were selected as the control 2 group.The expressions of IL-17 and FOXP3 in CAD lesions and normal skin tissues were detected by immunofluorescence assay in paraffin sections.Corresponding statistical methods were used for statistical analysis.Results:The percentages of Th1,Th2,Th17,Treg,Th1 and Th2 in peripheral blood in the experimental 1 group were(25.81±6.52)(42.79±9.66)(12.34±5.15)(8.32±3.06),respectively.The percentages of Th1,Th2,Th17 and Treg in peripheral blood in the health group were(12.59±2.69)(50.88±9.06)(12.74±5.09)(10.42±2.07),respectively.The ratios of Th1/Th2 and Th17/Treg in the experimental 2 group were 0.58[0.48,0.74]and 1.65[1.00,2.21],respectively.In the control 1 group,the ratios of Th1/Th2 and Th17/Treg were 0.26[0.21,0.31]and 1.23[0.82,1.45],respectively.The positive rates of IL-17 and FOXP3 in skin lesions in the experimental 2 group were(85.91±11.73)and 3.60[1.97,4.83],respectively.The positive rates of IL-17 and FOXP3 in skin lesions in the control 1 group were(89.85±8.18)and 1.32[0.85,2.79],respectively.Conclusion:Low expression of Treg and high expression of Th1 in peripheral blood and high expression of FOXP3 in skin lesions in CAD patients during non-acute attack may be related to the imbalance of Th17/Treg and Th1/Th2 expression in peripheral blood and IL-17/FOXP3 expression in skin lesions.There was no significant difference in Th17/Treg,Th17 percentage and IL-17 expression in skin lesions.
