Effects of the Dachengqi decoction on gastrointestinal function and inflammatory factors in rats with sepsis gastrointestinal dysfunction
Objective:To study the effects of the Dachengqi decoction(大承气汤)on gastrointestinal function and inflammatory factors in rats with septic gastrointestinal dysfunction.Methods:Fifty male Wistar rats were randomly divided into the sham-operated group,model group,the Dachengqi decoction high(10 g/kg),medium(5 g/kg)and low(2.5 g/kg)dose groups,with ten rats in each group.Except for sham-operated group,the rest were given cecal ligation and perforation to estabish septic gastrointestinal dysfunction models.The Dachenqi decoction groups were given corresponding dose of the Dachenqi decoction,the sham-operated group and the model group were given the equal amounts of physiological saline,were continuously gavaged once a day,for 7 days.After administration,the gastric residual rate and intestinal advancement rate of rats were measured by semi-solid paste method,and the serum levels of gastrointestinal hormones motilin,gastrin and inflammatory factors tumor necrosis factor-α and interleukin-1β were detected by enzyme-linked immunosorbent assay.Results:Compared with the sham-operated group,gastric emptying rate,intestinal propulsion rate,motilin and gastrin levels in the model group were significantly decreased,while tumor necrosis factor-α and interleukin-1β levels were significantly increased,with significant statistical differences(P<0.01).Compared with the model group,the gastric emptying rate and intestinal propulsion rate of rats in each dose groups of the Dachengqi decoction were increased to varying degrees,with significant statistical differences(P<0.01);the levels of motilin and gastrin in the high-dose and medium-dose groups of the Dachengqi decoction were significantly increased,with significant statistical differences(P<0.01).The level of tumor necrosis factor-α in each dose groups of the Dachengqi decoction were significantly decreased,and the differences were statistically significant(P<0.01),while the level of interleukin-1β in the high-dose and medium-dose groups of Dachengqi decoction were significantly decreased,and the differences were statistically significant(P<0.01).Conclusion:Dachengqi decoction could effectively promote gastrointestinal motility in rats with septic gastrointestinal dysfunction,increase the secretion level of gastrointestinal hormones,inhibit the release of inflammatory factors,and reduce the inflammatory reaction of sepsis.
