基于网络药理学研究清热调糖饮治疗湿热内蕴型2型糖尿病的作用机制

目的:运用网络药理学探讨清热调糖饮治疗湿热内蕴2型糖尿病的作用机制.方法:检索中药系统药理学数据库与分析平台,筛选组方的有效成分,并预测其潜在靶点.利用DisGenet检索2型糖尿病靶点,通过Venny软件获取药物有效成分作用靶点与疾病基因靶点的交集.利用STRING构建蛋白质-蛋白质相互作用网络,利用DAVID数据库进行基因本体论(GO)和京都基因与基因组百科全书(KEGG)通路富集分析.结果:清热调糖饮与2型糖尿病的共同基因靶点有133个,其中君药黄连可作用的疾病靶点占比最高,为86.47%;其次为玉米须、大黄;茯苓的作用靶点最少.经GO分析,生物过程共387条,细胞组成共40条,分子功能共74条,其中与胰岛素代谢相关的共6条,与内皮功能相关的共11条.KEGG分析筛选得到134条通路(P<0.01),其中基因数目≥10的通路有88条,与糖尿病相关的信号通路共7条,与糖尿病并发症相关的高富集信号通路共4条.结论:清热调糖饮配伍科学,可能通过晚期糖基化终末产物及其受体(Advanced Glycation End Products-Receptor Advanced Glycation End Products,AGE-RAGE)信号通路、核因子-κB(Nuclear Factor-κ B,NF-κB)信号通路和肿瘤坏死因子(Tumor Necrosis Factor,TNF)信号通路等实现对湿热内蕴2型糖尿病及并发症的治疗作用.
A study on the mechanism of treating type 2 diabetes of damp-heat type with the Qingre Tiaotang decoction based on network pharmacology
Objective:To explore the mechanism of the Qingre Tiaotang decoction(清热调糖饮)in the treatment of type 2 diabetes of damp-heat type by network pharmacology.Methods:Active pharmaceutical ingredients were screened by the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)database,and potential targets were predicted.Type 2 diabetes targets were required by DisGenet databases.The intersection of drug active ingredient targets and disease gene targets was obtained by Venny.Gene ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of genes and genomes(KEGG)pathways enrichment analysis were performed for potential targets by DAVID databases.Results:There were 133 common gene targets between the Qingre Tiaotang decoction and type 2 diabetes.The proportion of available targets Huanglian(Rhizoma Coptidis)affected was 86.47%,more than Yumixu(Stigma Maydis)and Dahuang(Radix et Rhizoma Rhei).The number of targets of Fuling(Poria)was lowest.A total of 387 terms of biological process,40 terms of cellular component and 74 terms of molecular function were obtained by GO enrichment analysis.Six terms were related to insulin metabolism,and eleven terms were related to endothelial function.KEGG pathways dressed 134 terms(P<0.01)by screening,including 88 terms with more than 10 genes.Among these items,there were seven signaling pathways associated with diabetes and four highly enriched pathways associated with diabetic complications.Conclusion:The compatibility of the Qingre Tiaotang decoction was reasonable,which owned therapeutic effects on type 2 diabetes of damp-heat type and complication through advanced glycation end products and their receptor(AGE-RAGE)signaling,nuclear factor Kb(NF-κB)signaling,and tumor necrosis factor(TNF)signaling.
