基于网络药理学研究川芎-赤芍治疗冠状动脉粥样硬化性心脏病心绞痛的作用机制

目的:通过网络药理学理念和技术研究川芎-赤芍药对治疗冠状动脉粥样硬化性心脏病(简称冠心病)心绞痛的主要活性成分、靶点及生物学过程和通路.方法:通过TCMSP数据库检索出川芎和赤芍的主要活性成分、靶点信息,再通过GeneCards、OMIM、TTD数据库获取冠心病心绞痛的相关靶点信息,继而得到川芎-赤芍与冠心病心绞痛的交集靶点,采用Cytoscape 3.10.0软件构建中药-成分-靶点-疾病靶点网络,将获取的交集靶点信息通过STRING数据库构建蛋白质-蛋白质相互作用网络,筛选出核心靶点.利用David数据库对交集靶点进行基因本体论(GO)功能富集分析和京都基因与基因组百科全书(KEGG)通路富集分析.结果:从川芎和赤芍中共筛选得到36个主要活性成分,其中川芎7个、赤芍29个,相关作用靶点共166个,筛选去重后得到76个,冠心病心绞痛的相关作用靶点共1213个,川芎和赤芍与冠心病心绞痛交集靶点共35个,核心靶点18个.GO功能富集分析结果显示主要涉及对外源刺激的反应、凋亡过程的负调控、细胞外区域、质膜、酶绑定、相同蛋白结合等生物过程,KEGG通路富集分析结果显示主要涉及癌症通路、流体剪切力与动脉粥样硬化、卡波西肉瘤相关疱疹病毒感染、癌症中的蛋白聚糖类、化学致癌作用-受体活化、血脂与动脉粥样硬化等信号通路.结论:本研究初步揭示了川芎-赤芍药对治疗冠心病心绞痛疾病的主要活性成分、靶点、生物学过程和通路,为进一步研究其药理学作用及临床应用提供参考依据.
A study on the mechanism of action of Chuanxiong-Chishao in the treatment of angina pectoris in coronary artery disease based on network pharmacology
Objective:To investigate the mechanism of action of Chuanxiong(Rhizoma Chuanxiong)and Chishao(Radix Paeoniae Rubra)in the treatment of angina pectoris in coronary heart disease(CHD-AP)through the concept and technology of network pharmacology.Methods:The TCMSP database was used to retrieve the main active ingredients and targets of Chuanxiong and Chishao,and then the GeneCards,OMIM and TTD databases were used to obtain the relevant target information of CHD-AP,followed by the intersection targets of Chuanxiong-Chishao and CHD-AP,and the Cytoscape 3.10.0 software was used to construct the network of Chinese medicine-ingredient-target-disease target.The intersection target information was then used to construct a protein-protein interaction(PPI)network by the STRING database to screen out the core targets.The intersection targets were enriched for GO function and KEGG pathway using the David database.Results:A total of 36 major active ingredients were screened from Chuanxiong and Chishao,including 7 from Chuanxiong and 29 from Chishao,and a total of 166 relevant action targets were obtained,after screening and de-weighting,there were 76 from CHD-AP,1 213 from CHD-AP,35 from Chuanxiong and Chishao and CHD-AP,with 18 core targets.The results of GO functional enrichment analysis showed that they mainly involved in biological processes such as response to exogenous stimuli,negative regulation of apoptotic process,extracellular regions,plasma membrane,enzyme binding,identical protein binding,etc..The results of KEGG pathway enrichment analysis showed that there were mainly signaling pathways such as cancer pathway,fluid shear and atherosclerosis,Kaposi's sarcoma-associated herpesvirus infection,proteoglycans in cancer,chemical carcinogenesis-receptor activation,lipids and atherosclerosis,etc..Conclusion:This study initially revealed the main active ingredients,targets,biological processes and pathways of Chuanxiong-Chishao in the treatment of CHD-AP,which provides a reference basis for further research on its pharmacological effects and its clinical application.
