Discussion on the mechanism of Huangqi-Danshen in treating coronary heart disease based on network pharmacology
Objective:This study was aimed to investigate the effective components,targets and related pathways of Huangqi(Radix Astragali)-Danshen(Radix et Rhizoma Salviae Miltiorrhizae)in the treatment of coronary atherosclerotic heart disease through network pharmacology.Methods:The main components and action targets of Huangqi-Danshen were searched.Traditional Chinese medicine systems pharmacology database and analysis platform was adopted to screen the active ingredients and targets of Huangqi-Danshen.Human gene databases GeneGards and DrugBank were used to search CHD related targets.The common target of drugs and diseases were selected to draw Venn diagram.The common targets of drugs and diseases were analyzed by STRING online analysis platform,and the protein-protein interaction network was constructed.The Metascape database was used for gene ontology(GO)functional enrichment analysis of common drug and disease targets,and the Kyoto encyclopedia of genes and genomes genomes(KEGG)pathway enrichment analysis was conducted.The Cytocsape 3.9.1 software was used to construct the TCM-effective component-target-disease-pathway network diagram.Results:A total of 85 effective chemical components were selected from Huangqi-Danshen,including kaempferol,quercetin,tanshinone ⅡA,etc.,and 100 targets were associated with coronary heart disease.The results of GO analysis showed 672 biological functions,4 753 biological processes and 672 cytological components.KEGG results showed that 184 signaling pathways were obtained to play a role in the treatment of coronary heart disease.Conclusion:This study explored the possibility of Huangqi-Danshen participating in the treatment of coronary heart disease through multi-component,multi-target and multi-pathway,providing a new theoretical basis for subsequent research.
