摘要

目的:通过网络药理学预测搜风化痰方治疗心房颤动的作用靶点及信号通路,进一步探讨其治疗心房颤动的潜在作用机制.方法:用中药系统药理学数据库与分析平台(TCMSP)筛选搜风化痰中药复方的活性成分并预测其作用靶点,构建活性成分-预测靶点网络.通过OMIM、TTD、GeneCards检索心房颤动的潜在靶点,采用STRING 11.0获取心房颤动靶点蛋白质相互作用,通过CytoScape 3.9.1软件对搜集到的数据进行可视化分析,进而构建心房颤动靶点的蛋白质-蛋白质相互作用网络.通过Venny将搜风化痰中药复方预测靶点与心房颤动靶点进行合并,得到搜风化痰中药复方治疗心房颤动的潜在靶点,运用DAVID数据库对搜风化痰方治疗心房颤动的潜在靶点进行生物功能及信号通路富集分析.通过微生信平台进行数据可视化分析,CytoScape 3.9.1软件构建搜风化痰方治疗心房颤动的活性成分-潜在靶点-信号通路网络.结果:预测出搜风化痰中药复方治疗心房颤动的潜在靶点25个,中药复方可通过糖尿病并发症中的晚期糖基化终末产物-晚期糖基化终末产物受体信号通路、松弛素信号通路、白细胞介素-17信号通路等发挥治疗心房颤动的作用.结论:搜风化痰中药复方基于多成分、多靶点、多通路治疗心房颤动.

Abstract

Objective:To predict the targets and signaling pathways of the removing wind and reducing phlegm prescription for the treatment of atrial fibrillation through network pharmacology,so as to further explore the potential mechanism of action on atrial fibrillation.Methods:The TCMSP was used to screen the active ingredients and predict the targets of the removing wind and reducing phlegm prescription,and the active ingredient-target network was constructed.The potential targets of atrial fibrillation were searched by OMIM,TTD and GeneCards,and the protein interaction of atrial fibrillation was obtained by STRING 11.0.The data was visualized and analyzed by CytoScape 3.9.1 to construct protein-protein interaction networks of atrial fibrillation targets.Through Venny,the predicted targets of the removing wind and reducing phlegm prescription were merged with the atrial fibrillation targets to obtain the potential targets of the removing wind and reducing phlegm prescription for the treatment of atrial fibrillation,and then the database DAVID was utilized to analyze the biofunctionality and signaling pathways of the potential targets of the removing wind and reducing phlegm prescription for the treatment of atrial fibrillation.Data visualization and analysis were carried out through Bioinformatics platform,and CytoScape 3.9.1 software was used to construct the active ingredient-potential target-signaling pathway network of the removing wind and reducing phlegm prescription for the treatment of atrial fibrillation.Results:Twenty-five potential targets were predicted for the treatment of atrial fibrillation with the removing wind and reducing phlegm prescription,which could play a role in the treatment of atrial fibrillation through the AGE-RAGE signaling pathway in the diabetic complication,relaxin signaling pathway and IL-17 signaling pathway.Conclusion:The Removing wind and reducing phlegm prescription is based on multi-components,multi-targets and multi-pathways to treat atrial fibrillation.
