基于网络药理学及质谱技术探讨桃核承气汤治疗盆腔瘀血综合征的作用机制研究

目的:结合超高效液相飞行时间高分辨质谱联用系统(Xevo G2-XS QTOF MS)和网络药理学预测桃核承气汤治疗盆腔瘀血综合征(Pelvic Congestion Syndrome,PCS)的作用靶点及潜在作用机制.方法:根据《伤寒论》记载,制备桃核承气汤水煎剂,并采用Xevo G2-XS QTOF MS对其化学成分进行解析,根据所得精确相对分子质量、二级质谱裂解碎片等质谱信息,结合从TCMSP等文献数据库获取的各药味的化学成分,利用SwissADME数据库通过药代动力学与类药性等参数筛选出各药味的活性成分,再通过SwissTargetPrediction数据库以可能性≥ 0.5为标准,筛选出活性成分相应的靶点,建立"药材-成分-靶点"的分子网络,以"pelvic congestion syndrome"作为关键词,检索O MIM、GeneCards、GrugBank等疾病数据库,以其相关性得分的中位数为标准筛选出疾病关键靶点.基于药物与疾病的共同靶点,利用STRING数据库进行蛋白质-蛋白质相互作用分析,选取置信度高于0.9的数据,导入Cytoscape软件进行网络拓扑分析,筛选出度值前5的核心成分与度值前10的核心靶点,将核心靶点导入Metascape数据库进行基因本体论(GO)与京都基因与基因组百科全书(KEGG)富集分析.最后采用AutoDock Vina软件对核心成分与核心靶点进行分子对接.结果:最终获得42个活性成分与137个相关药物靶点,1251个疾病靶点,39个共同靶点.经GO与KEGG分析,获得生物过程条目669个、细胞组分条目26个、分子功能条目62个,以及KEGG信号通路164条.分子对接结果显示,槲皮素、山柰酚、白杨素等5个核心成分(度值前5)与肿瘤坏死因子(Tumor Necrosis Factor,TNF)、Akt丝氨酸/苏氨酸蛋白激酶1(Akt serine/threonine kinase 1,AKT1)、肉瘤基因(Sarcoma,SRC)、表皮生长因子受体(Epidermal Growth Factor Receptor,EGFR)等15个核心靶点(度值前15)均有较好的结合活性.结论:桃核承气汤可能通过调控表皮生长因子受体络氨酸激酶抑制剂(Epidermal Growth Factor Receptor-Tyrosine kinase inhibitor,EGFR-TKI)耐药性、雌激素、磷脂酰肌醇 3 激酶-蛋白激酶 B(Phosphatidylinositol 3 kinase-protein kinase B,PI3K-Akt)、Ras 相关蛋白 1(Ras-proximate-1,RAP 1)、大鼠肉瘤(Rat Sarcoma,RAS)和催乳激素等信号通路来促进新生血管形成与破损血管修复、扩张血管、调节血管舒张,从而降低血压、改善血液粘度以治疗PCS,为后续进一步研究桃核承气汤治疗瘀血证的药效物质和作用机制提供参考.
A study on the mechanism of treating pelvic congestion syndrome with the Taohe Chengqi decoction based on network pharmacology and mass spectrometry
Objective:To predict the target and potential mechanism of treating pelvic congestion syndrome(PCS)with the Taohe Chengqi decoction(桃核承气汤)by ultra-performance liquid time-of-flight high resolution mass spectrometry(Xevo G2-XS QTOF MS)and network pharmacology.Methods:According to the records in Shanghan Lun(《伤寒论》),the Taohe Chengqi decoction was prepared,and its chemical composition was analyzed using the Xevo G2-XS QTOF MS.The chemical composition of each drug was obtained according to the obtained precise molecular weight,secondary mass spectrometry cracking fragment and other mass spectrometry information,combined with the literature in database such as TCMSP.SwissADME database was used to screen the active ingredients of each drug taste through pharmacokinetic and drug-like parameters,and then SwissTargetPrediction database was used to screen the corresponding target of active ingredients with the possibility not less than 0.5 as the standard,and a molecular network of medicinal material-ingredient-target was established.Lising pelvic congestion syndrome as a keyword,the median of correlation scores was used to search disease databases(OMIM,GeneCards,GrugBank),and the key disease targets were screened out.Based on the common targets of drugs and diseases,the STRING database was used for PPI analysis.Data with a confidence level higher than 0.9 was selected and imported into Cytoscape software for network topology analysis to screen out the top 5 core components and the top 10 core targets.The core targets were imported into Metascape database for GO and KEGG analysis.Finally,AutoDock Vina software was used to perform molecular docking between core components and core targets.Results:Forty-two active ingredients and 137 related drug targets,1 251 disease targets and 39 common targets were obtained.By GO and KEGG analyses,669 items of biological process,26 items of cell component,62 items of molecular function and 164 signaling pathways of KEGG were obtained.The molecular docking results showed that five core components(top 5 in degree value)including quercetin,kaferol and chrysin had good binding activities with 15 core targets(top 15 in degree value)including TNF,AKT1,SRC and EGFR.Conclusion:The Taohe Chengqi decoction may regulate EGFR-TKI resistance,estrogen,PI3K-Akt,RAP1,RAS,prolactin and other signaling pathways to promote the formation of new blood vessels,repair damaged blood vessels,dilate blood vessels,regulate blood vessel dilation,thereby reducing blood pressure and improving blood viscosity to treat PCS.It provides reference for further study on the pharmacodynamic substances and action mechanism of the Taohe Chengqi decoction in treating blood stasis syndrome.
