The application of the Buyang Huanwu decoction in promoting astrocyte proliferation in the peripheral tissue of cerebral ischemia/reperfusion rats
Objective:To observe whether the Buyang Huanwu decoction(补阳还五汤dintothdivided)can promote astrocyte proliferation in cere-bral ischemia/reperfusion rats.Methods:Fifty-four SD rats were randomly divided into the sham operation group,the model group and the Buyang Huanwu decoction group,with 18 rats in each group.Each group was divided into 3 subgroups according to the reperfusion time of 1 day,3 days and 7 days.Except for the sham operation group,the other groups were treated by middle cerebral artery occlusion(MCAO)model induced by thread embolism and reperfusion after 90 min of ischemia.Twenty four hours after modeling,the Buyang Huanwu decoction group was given the Buyang Huanwu decoction by gavage(12.72 g/kg per day,divided into two doses),the sham operation group and the model group were given the same amount of physiological saline by gavage until the day before sacrifice.Immunofluorescence staining was used to detect GFAP positive cells and evaluate the number of astrocytes.Results:Compared with the sham operation group,the number of astrocytes in the peripheral tissue of ischemic area was decreased significantly in the model group 3 and 7 days after cerebral ischemia/reperfusion(P＜0.01).Compared with the model group,the number of astrocytes in the Buyang Huanwu decoction group was increased significantly 3 and 7 days after cerebral ischemia/reperfusion(P＜0.01).Conclusion:The Buyang Huanwu decoction can promote astrocyte proliferation in the peripheral tissue of cerebral ischemia/reperfusion rats.The specific mechanism needs to be further studied.
