补阳还五汤促进脑缺血/再灌注大鼠缺血区周边组织星型胶质细胞增生

目的:观察补阳还五汤是否可以促进脑缺血/再灌注大鼠星型胶质细胞增生.方法:将54只SD大鼠随机分为假手术组、模型组、补阳还五汤组,每组18只,每组又按再灌注时间分为1 d、3d、7 d共3个亚组.除假手术组外,其余各组采用线栓法诱导大脑中动脉阻塞(Middle Cerebral Artery Occlusion,MCAO)模型,缺血90 min后再灌注.建模后24 h补阳还五汤组开始给予补阳还五汤灌胃(每天12.72 g/kg,分2次给药),假手术组、模型组给予等量生理盐水灌胃,直至处死的前一天.免疫荧光染色法检测胶质纤维酸性蛋白(Glial Fibrillary Acidic Protein,GFAP)阳性细胞,评估星型胶质细胞数量.结果:与假手术组比较,模型组脑缺血/再灌注后3d、7d缺血区周边组织星型胶质细胞数量明显减少(P<0.01);与模型组比较,补阳还五汤组脑缺血/再灌注后3 d、7 d,缺血区周边组织星型胶质细胞数量明显增多(P<0.01).结论:补阳还五汤可以促进脑缺血/再灌注大鼠缺血区周边组织星型胶质细胞增生,具体机制尚需进一步研究.
The application of the Buyang Huanwu decoction in promoting astrocyte proliferation in the peripheral tissue of cerebral ischemia/reperfusion rats
Objective:To observe whether the Buyang Huanwu decoction(补阳还五汤dintothdivided)can promote astrocyte proliferation in cere-bral ischemia/reperfusion rats.Methods:Fifty-four SD rats were randomly divided into the sham operation group,the model group and the Buyang Huanwu decoction group,with 18 rats in each group.Each group was divided into 3 subgroups according to the reperfusion time of 1 day,3 days and 7 days.Except for the sham operation group,the other groups were treated by middle cerebral artery occlusion(MCAO)model induced by thread embolism and reperfusion after 90 min of ischemia.Twenty four hours after modeling,the Buyang Huanwu decoction group was given the Buyang Huanwu decoction by gavage(12.72 g/kg per day,divided into two doses),the sham operation group and the model group were given the same amount of physiological saline by gavage until the day before sacrifice.Immunofluorescence staining was used to detect GFAP positive cells and evaluate the number of astrocytes.Results:Compared with the sham operation group,the number of astrocytes in the peripheral tissue of ischemic area was decreased significantly in the model group 3 and 7 days after cerebral ischemia/reperfusion(P<0.01).Compared with the model group,the number of astrocytes in the Buyang Huanwu decoction group was increased significantly 3 and 7 days after cerebral ischemia/reperfusion(P<0.01).Conclusion:The Buyang Huanwu decoction can promote astrocyte proliferation in the peripheral tissue of cerebral ischemia/reperfusion rats.The specific mechanism needs to be further studied.
