白斑冲剂含药血清对过氧化氢诱导的人表皮黑素细胞氧化损伤的保护作用

目的:研究白斑冲剂含药血清对过氧化氢(H2O2)诱导的正常人表皮黑素细胞氧化损伤的保护作用.方法:以不同浓度H2O2处理正常人表皮黑素细胞PIG1,采用CCK-8法检测PIG1细胞的活力,选取H2O2最佳作用浓度,建立体外黑素细胞氧化应激模型.以不同浓度白斑冲剂含药血清和不同作用时间干预黑素细胞氧化应激模型,CCK-8法筛选含药血清合适的浓度和作用时间.将对数生长期的PIG1细胞随机分为对照组(PIG1细胞+正常大鼠血清)、模型组(PIG1细胞+H2O2)、白斑冲剂组(PIG1细胞+白斑冲剂含药血清+H2O2).干预后,倒置显微镜下观察白斑冲剂含药血清对PIG1细胞氧化损伤的保护作用,采用流式细胞术检测各组PIG1细胞凋亡.结果:H2O2作用下PIG1细胞的活性呈浓度依赖性降低,依据ID50值,选取0.8 mmol/L H2O2作用24 h建立PIG1细胞的氧化应激模型.氧化应激下PIG1细胞在不同浓度含药血清0 h、12 h、24 h、48 h、72 h作用下,与模型组比较,20%含药血清作用24 h细胞活力升高最为显著,因此选择20%浓度的白斑冲剂血清处理24 h作为含药血清的最佳浓度和作用时间.倒置显微镜下结果显示,与对照组比较,模型组细胞密度减少,死细胞数量增加.白斑冲剂组与模型组相比,细胞损伤程度明显减弱.与对照组比较,模型组细胞凋亡比例明显上升(P<0.01);与模型组比较,白斑冲剂组细胞凋亡比例明显下降(P<0.01).结论:白斑冲剂含药血清对H2O2诱导的正常人黑素细胞氧化损伤具有明显的保护作用.
The protective effect of serum containing Baiban Granules on oxidative damage of human epidermal melanocytes induced by H2O2
Objective:To study the protective effect of serum containing Baiban Granules(白斑冲剂)on the oxidative damage of normal epidermal melanocytes induced by hydrogen peroxide(H2O2).Methods:Normal epidermal melanocytes PIG1 were treated with different con-centrations of H2O2,the activity of PIG1 cells was detected by CCK-8 method,and the optimal concentration of H2O2 was selected to establish the oxidative stress model of melanocytes in vitro.Different concentrations of Baiban Granules containing serum and action time were used to interfere with melanocyte oxidative stress model.CCK-8 method was used to screen the appropriate concentration and action time of Baiban Granules.PIG1 cells at logarithmic growth stage were randomly divided into the control group(PIG1 cells+normal rat serum),the model group(PIG1 cells+H2O2),and Bai-ban Granules group(PIG1 cells+Baiban Granules drug-containing serum+H2O2).The protective effect of Baiban Granule on oxidative damage of PIG1 cells was observed under inverted microscope after intervention.PIG1 cell apoptosis was detected by flow cytometry.Results:The activity of PIG1 cells was decreased in a concentration-dependent manner under the effect of H2O2.The oxidative stress model of PIG1 cells was established by 0.8 mmol/L H2O2 for 24 h according to ID50 value.Under oxidative stress,PIG1 cells were treated with different concentrations of drug-containing serum for 0 h,12 h,24 h,48 h and 72 h.Compared with the model group,20%drug-containing senim showed the most significant increase in cell viability for 24 h.Therefore,20%Baiban Granules serum was selected as the optimal concentration and action time of drug-containing serum for 24 hours.The inverted microscope showed that the cell density was decreased and the number of dead cells was increased in the model group compared with those in the control group.Compared with the model group,the degree of cell damage in Baiban Granules group was significantly reduced.Compared with the control group,the apoptosis ratio in the model group was significantly increased(P<0.01).Compared with the model group,the proportion of apoptosis in Baiban Granules group was significantly decreased(P<0.01).Conclusion:The serum containing Baiban Granules has an obvious protective effect on the oxidative damage of normal melanocytes induced by H2O2.
