目的:通过生物信息学方法,探讨中性粒细胞外诱捕网(Neutrophil Extracellular Traps,NETs)相关基因在呼吸道合胞病毒(Respiratory Syncytial Virus,RSV)感染患儿全血中的表达模式,并预测潜在中药.方法:根据NETs相关基因表达情况对数据集进行无监督聚类分析,对组间进行基因集富集分析(GSEA)及免疫浸润分析,通过差异分析鉴定组间NETs相关差异基因(NETsDEGs),借用CTD数据库及Coremine Medical数据库进行中药预测.结果:根据NETs相关基因的表达水平,RSV感染患儿被分为两组,GSEA富集分析获得的多条通路与免疫相关,免疫浸润分析表明两组之间免疫细胞浸润丰度存在差异,进一步提示NETs水平对机体免疫环境的影响,差异分析鉴定髓过氧化物酶(Myeloperoxidase,MPO)、天青杀素1(Recombinant Azurocidin 1,AZU1)、组织蛋白酶G(Cathepsin G,CTSG)和中性粒细胞弹性蛋白酶(Elastase,Neutrophil Expressed,ELANE)为两组间NETsDEGs,据此共映射到相关化合物46种,获得预测中药145味,对两组共同的预测中药进行分析,发现NETs在RSV感染中的病位主要在肺脾两脏,预测中药的性味以苦、甘、辛、寒为主,预示NETs在RSV感染中可能与风邪相关;此外对两组相应的预测中药进行比较,发现NETs表达水平与RSV感染证型及病位有关,其表达增加可能与热证相关.结论:本研究通过生物信息学方法,发现儿童RSV感染中NETs相关基因的两种表达情况,并据此预测了相关中药,通过对潜在中药的分析,丰富了儿童RSV感染的致病机制,对NETs的中医内涵进行了分析,为RSV感染的治疗提供了新的靶点.
Analysis of gene expression profiles related to NETs in children with RSV infection and prediction of Chinese medicine
Objective:To investigate the expression patterns of NETs-related genes in whole blood of RSV-infected children by bioinformatics methods and to predict potential Chinese medicines against NETs during RSV infection.Methods:According to the expression of NETs related genes,unsupervised cluster analysis was performed on the dataset,GSEA enrichment analysis and immune infiltration analysis were performed among the groups,and NETsDEGs among the groups were identified by difference analysis.CTD database and Coremine Medical database were used for TCM prediction.Results:According to the expression level of NETs related genes,RSV infected children were divided into two groups.Multiple pathways obtained by GSEA enrichment analysis were associated with immunity,and immunoinfiltration analysis showed that there were differences in the abundance of immune cell infiltration between the two groups,which further suggested the influence of NETs level on the immune environment of the body.MPO,AZU1,CTSG and ELANE were identified as NETsDEGs between the two groups by differential analysis,and a total of 46 related compounds were mapped accordingly,and 145 predicted Chinese medicines were obtained.The common predicted Chinese medicines of the two groups were analyzed,and it was found that the disease sites of NETs in RSV infection were mainly in the lung and spleen.It was predicted that the property and taste of traditional Chinese medicine was mainly bitter,sweet,pungent and cold,indicating that NETs might be related to wind evil in RSV infection.In addition,the two groups of corresponding predicted Chinese medicines were compared,and it was found that the expression level of NETs was related to the type and location of RSV infection,and its increase may be related to heat syndrome.Conclusion:In this study,two expressions of NETs-related genes in RSV-infected children are found through bioinformatics methods,and related Chinese medicines are predicted accordingly.Through the analysis of potential Chinese medicines,the pathogenic mechanism of RSV infection in children is enriched,and the connotation of NETs in Chinese medicine is analyzed,providing a new target for the treatment of RSV infection.
