A study on medication rule and mechanism of TCM in the treatment of perimenopausal vasomotor symptoms based on data mining and network pharmacology
Objective:To explore the medication rules of traditional Chinese medicine(TCM)in the treatment of vasomotor symptoms in perimenopausal period based on data mining and network pharmacology,and study the mechanism of core component.Method:The literature related to disease was collected from CNKI,VIP,and SinoMed since the establishment of the database.Microsoft Excel 2016,SPSS Statistics 22.0,Cytoscape software were used to analyze the medication rules of TCM compounds in the included literature.The intersection targets of TCM and diseases were obtained by TCMSP,GeneCards,DisGeNET and TTD databases.PPI network was constructed,GO and KEGG enrichment analyses were performed,and molecular docking was verified by Autodock.Results:A total of 26 literatures,42 TCM prescriptions and 79 TCM flavors were selected,and 4 core components were obtained.After analyzing the core components of"Coptidis rhizoma,Scutellariae radix,Radix Rehmanniae Praeparata,Angelica sinensis,Phellodendri chinensis cortex,Rehmanniae radix and Astragali radix",namely Danggui Liuhuang decoction(当归六黄汤),a total of 73 active ingredients,238 effective targets,512 disease targets,and 69 intersecting targets were obtained.The main active ingredients were quercetin,β-sitosterol,stigmasterol,etc..The core targets were IL-6,TNF,IL-1β,etc..It involved 20 BP items,14 CC items,18 MF items,and 20 KEGG biological pathway items.Molecular docking results verified the spontaneous binding and strong interaction between active components and core targets.Conclusion:Through data mining and network pharmacology,it is known that the treatment of Perimeno pausal vasomotor symptoms should mainly focus on tonifying and nourishing the kidneys,and clearing heat and reducing fire.The core component of Danggui Liuhuang decoction can play its role through multiple components,multiple targets and multiple pathways.
