基于网络药理学和数据挖掘探讨中药调控铁死亡干预银屑病的用药规律

目的:运用网络药理学及数据挖掘技术探讨中药调控铁死亡干预银屑病的用药规律,为银屑病相关研究提供参考.方法:利用GeneCards、GeneCLiP3和FerrDb数据库获取铁死亡的强相关靶点,通过GeneCards和DisGeNET数据库获取银屑病疾病靶点,取两者交集,得到银屑病铁死亡的相关靶点.对银屑病铁死亡靶点进行蛋白质-蛋白质相互作用分析,选取度值≥2倍中位数的靶点,通过中药系统药理学数据库与分析平台(TCMSP)获取相关化合物和中药.利用Cytoscape 3.10.1软件构建潜在靶点-化合物网络图、靶点-化合物-中药网络图.结果:获得银屑病铁死亡相关靶点227个,度值≥108的银屑病铁死亡潜在靶点48个,筛选得到37个化合物和784味中药.结论:从银屑病和铁死亡的靶点出发,挖掘通过调控铁死亡干预银屑病的中药及用药规律,探讨银屑病铁死亡的相关作用机制,为银屑病铁死亡的相关研究提供参考.
The medication rule of TCM medicine in treating psoriasis by regulating ferroptosis based on network pharmacology and data mining technology
Objective:In this paper,network pharmacology and data mining technology was mainly used to explore the medication rule of TCM medicine on psoriasis by regulating ferroptosis,so as to provide ideas for the related research on psoriasis.Methods:The GeneCards,GeneCLiP3 and FerrDb databases were used to obtain the strong related targets of ferroptosis,and GeneCards and DisGeNET databases were used to gain the psoriasis disease targets.The intersection of the two was taken to obtain the relevant targets of ferroptosis in psoriasis.The protein-protein interaction analysis was performed on the target of ferroptosis in psoriasis,and the targets with degree values of not less than 2 times the median were selected.Related compounds and TCM medicine were obtained by TCMSP.Cytoscape 3.10.1 software was used to build potential target-compound network diagram,and target-compound-TCM medicine network diagram.Results:A total of 227 targets related to ferroptosis in psoriasis were obtained,and 48 potential targets with degree values of no less than 108 were identified.A total of 37 compounds and 784 kinds of TCM medicine were screened.Conclusion:Starting from the targets of psoriasis and ferroptosis,this study explores the TCM medicine and the medication rule in regulating ferroptosis in psoriasis,and the related action mechanism of TCM medicine on ferroptosis in psoriasis,which can provide reference for the related research of ferroptosis in psoriasis.
