金龙降尿酸颗粒治疗高尿酸血症的作用机制研究

目的:基于网络药理学与分子对接方法探讨金龙降尿酸颗粒治疗高尿酸血症的作用机制。方法:通过中药系统药理学数据库与分析平台(TCMSP)获取金龙降尿酸颗粒的主要活性成分及靶点,通过OMIM、DrugBank、GeneCards等数据库获取高尿酸血症的靶点,利用Venny 2。1。0得到中药与疾病的交集靶点,利用Cytoscape 3。10。0绘制"中药-活性成分-治疗靶点"网络图,利用STRING数据库及Cytoscape 3。10。0绘制蛋白质-蛋白质相互作用网络图,利用DAVID数据库进行富集分析,通过CB-Dock2进行分子对接验证。结果:搜集到金龙降尿酸颗粒43个活性成分、248个靶点,获得了高尿酸血症1 056个疾病靶点,其中中药与疾病的交集靶点共有91个;获得了基因本体论(GO)功能富集分析条目531个,京都基因与基因组百科全书(KEGG)通路143条;分子对接结果显示关键成分与关键靶点均具有良好的结合能力。结论:金龙降尿酸颗粒通过调节白细胞介素(Interleukin,IL)-6、肿瘤坏死因子(Tumor Necrosis Factor,TNF)、肿瘤蛋白p53等靶点抑制晚期糖基化终末产物-晚期糖基化终末产物受体(Advanced Glycation End Product-Receptor for Advanced Glycation End Product,AGE-RAGE)、IL-17、TNF 等信号通路,起到对高尿酸血症的治疗作用。
A study on the action mechanism of the Jinlong Jiangniaosuan granules on hyperuricemia
Oobjective:To investigate the action mechanism of the Jinlong Jiangniaosuan granules(金龙降尿酸颗粒)on hyperuricemia based on network pharmacology and molecular docking.Methods:The main active ingredients and targets of the Jinlong Jiangniaosuan granules were obtained from TCMSP database.Targets of hyperuricemia were searched through OMIM,DrugBank,GeneCards and other databases.The intersection target of chinese medicine and disease was obtained by Venny 2.1.0.The Chinese medicine-active ingredient-therapeutic target network was mapped by Cytoscape 3.10.0.Protein-protein interaction network was mapped by STRING database and Cytoscape 3.10.0.Enrichment analysis was made by the DAVID database.Molecular docking validation was made by CB-Dock2.Results:Forty-three active ingredients and 248 targets of the Jinlong Jiangniaosuan granules were collected.A total of 1 056 disease targets of hyperuricemia were obtained.There were a total of 91 intersection targets of Chinese medicine and disease.A total of 531 entries of GO functional enrichment analysis and 143 entries of KEGG pathway enrichment analysis were obtained.The molecular docking results showed that the key components and the key targets all had good binding ability.Conclusion:The Jinlong Jiangniaosuan granules play a therapeutic role in treating hyperuricemia by regulating the targets of IL-6,TNF and TP53,and inhibiting the signaling pathways such as AGE-RAGE,IL-17 and TNF.
