Objective To compare the contents of effective constituents of gardenoside, crocin- I and crocin- II in the fruits, seeds and pericarp of Gardeniajasminoides from different producing places. Methods Wavelength conversion RP-HPLC method was used. A Thermo ODS C18(250 mm × 4.6 mm, 5μm)column was used with the mobile phase consisting of acetonitrile-0.1%phosphoric acid( gradient elution) at the flow rate of 1.0 mL·min-1. The UV detection wavelength was set at 238 nm for gardenoside and 440 nm for crocin- I and crocin- II . Results The measured peaks were completely separated from the peaks of other components. The linear ranges of gardenoside, crocin-I and crocin-II were 0.420 ~ 5.252 μg(r=0.9993), 0.199 ~ 2.495 μg(r=0.9998) and 0.023 ~ 0.288 μg (r=0.9997), respectively. The average recovery rates of gardenoside, crocin- I and crocin-II were 98.23%(RSD=0.52%,n=6), 97.88%(RSD= 2.02% ,n -6) and 97.77% (RSD =1.47% ,n =6), respectively. Conclusion The method is sample, accurate and reproducible for the effective constituent contents in different positions of Gardenia jasminoides from different producing places.
