白蔹炮制前后指纹图谱的建立和比较研究

摘要
目的 比较白蔹炮制前后成分含量的变化,建立白蔹炮制前后指纹图谱质量控制方法.方法 采用HPLC法,在检测波长290 nm,流速1.0 mL·min-1的条件下,以乙腈-0.2%磷酸水为流动相进行梯度洗脱,建立不同批次白蔹炮制前后的HPLC指纹图谱,并运用中药色谱指纹图谱相似度评价系统进行数据分析;同时用HPLC法测定白蔹生品和炮制品中没食子酸、 原儿茶酸、 原儿茶醛、 白藜芦醇的含量.结果 以不同批次的白蔹炮制前后获得的8个共有峰为特征峰,建立白蔹药材的指纹图谱.含量测定结果显示,白蔹炮制品中的原儿茶酸、 没食子酸的含量明显高于生品,原儿茶醛、 白藜芦醇的含量略低于生品.结论 白蔹样品指纹图谱及指标成分含量测定可用于控制白蔹药材的质量.
Abstract
Objective To determine the changes of chemical components in Ampelopsis radix before and after processing,establish the quality control method of the fingerprint map of Ampelopsis Radix before and after processing. Methods HPLC was used under the condition of a detection wave length at 290 nm,a flow rate of 1.0 mL·min-1 and acetonitrile -0.2% phosphoric acid as the mobile phase gradient elution to detect the different batches of Ampelopsis radix before and after processing. Similarity evaluation system for chromatographic fingerprint of TCM was applied to analyze data. Meanwhile,the HPLC quantification for contents of gallic acid,protocatechuic acid,protocatechuic aldehyde and resveratrol was verified in Ampelopsis radix before and after processing.Results Eight common peaks are obtained as characteristic chromatographic peaks from different batches of Ampelopsis radix before and after processing, and the fingerprint of Ampelopsis radix was established. The determination results showed that in the processed products of Ampelopsis radix,protocatechuic acid and gallic acid contents were significantly higher than those in raw materials;the content of protocatechuic aldehyde,resveratrol were slightly lower than those in the crude Ampelopsis radix.Conclusion The HPLC fingerprints and the contents of the target components of Ampelopsis radix could be used to control the quality of Ampelopsis radix.
