目的 以牙龈卟啉单胞菌(porphyromonasgingivalis,Pg)和具核梭杆菌(fusobacteriumnucleatum,Fn)为供试菌株,研究桂花提取物槲皮苷的体外抑菌功能,以期为牙周相关疾病药物的开发提供有效的目标化合物.方法 通过液体培养基稀释法测定槲皮苷对两种供试菌株的最低抑菌浓度值(minimum inhibitory concentrations,MIC)和最低杀菌浓度值(minimum bactericidal concentration,MBC);结晶紫染色法(crystal violet staining solution,CVSS)测定槲皮苷对两种供试菌株生物膜形成的影响.通过噻唑蓝比色法(methyl thiazolyl tetrazolium,MTT)测定槲皮苷对两种供试菌株生物膜代谢活性的影响.结果 槲皮苷对两种供试菌株Pg和Fn的MIC分别为0.06和0.03 mg/mL,MBC分别为0.24和0.12 mg/mL.槲皮苷的浓度为0.03 mg/mL时,对Pg和Fn生物膜的形成和代谢活性均能产生明显的抑制作用.槲皮苷浓度达到0.24 mg/mL时,对两种菌株生物膜的形成和生物膜代谢活性抑制作用均达到85%以上.结论 槲皮苷对牙周常见致病菌Pg和Fn的体外生长均具有较强的抑制作用,有望成为一种安全有效的牙周抗菌新药.
Study on bacteriostasis of quercetin from the extract of osmanthus fragrans leaves on Pg and Fn in vitro
Objective To explore the antibacterial activity of quercetin from the extract of osmanthus fragrans leaves on porphyromonas gingivalis(Pg)and fusobacterium nucleatum(Fn)in vitro,in order to provide effective target compounds for the development of drugs for periodontal related diseases.Methods The minimum inhibitory concentration(MIC)and minimal bactericidal concentration(MBC)of quercetin to Pg and Fn were determined by broth microdilution assay.The effects of quercetin on the formation and metabolic activity of Pg and Fn biofilm were detected by crystal violet staining solution(CVSS)and methyl thiazolyl tetrazolium(MTT).Results The MIC and MBC of quercetin to Pg and Fn were 0.06 and 0.03 mg/mL,respectively,and 0.24 and 0.12 mg/mL,respectively.The formation and metabolic activity of Pg and Fn biofilm were inhibited by quercetin of 0.03 mg/mLQuercetin of 0.24 mg/mL inhibited Pg and Fn biofilm formation and metabolic activity by 85%.Conclusion Quercetin has a strong inhibitory effect on the in vitro growth of common periodontal pathogens Pg and Fn,and is expected to become a safe and effective new periodontal antibacterial drug.
