Improvement of Paeoniflorin on Endometrial Receptivity in Rats with Polycystic Ovary Syndrome and Influence on Wnt/β-catenin Pathway
Objective:To investigate the improvement effect and underlying mechanism of paeoniflorin on uterine endometrial receptivity in polycys-tic ovary syndrome(PCOS)rats.Methods:A PCOS model was induced in 40 female rats and the model rats were randomly divided into a model control group,low-and high-dose paeoniflorin groups(50 and 100 mg/kg),and a positive control group(Diane-35,350 mg/kg),with 10 rats in each group.Another 10 rats were assigned to the normal control group.Each group received the corresponding drugs orally for 21 consecutive days,and the normal control and the model control groups received an equal volume of saline.ELISA was used to measure the se-rum levels of estradiol(E2),testosterone(T),follicle-stimulating hormone(FSH),and luteinizing hormone(LH),as well as tumor necrosis factor-α(TNF-α),interleukin-1 β(IL-1β),and IL-6 in uterine endometrial tissues.Hematoxylin and eosin(HE)staining was performed to observe pathological changes in uterine endometrial tissues and to calculate and analyze the number of uterine endometrial glands and endome-trial thickness.RT-qPCR was used to detect the mRNA expression of Wnt3a and β-catenin in uterine endometrial tissues,and Western blot was employed to determine the protein expression of Wnt3a,β-catenin,estrogen receptor(ER),and progesterone receptor(PR)in uterine endometrial tissues.Results:Compared with the normal control group,the model control group exhibited significantly decreased E2,FSH,uter-ine endometrial thickness,and the number of uterine endometrial glands,significantly downregulated Wnt3a and β-catenin mRNA and protein expression,significantly elevated T,LH,TNF-α,IL-1β,IL-6 levels,and markedly upregulated ER and PR protein expression(P<0.05).Compared with the model control group,the paeoniflorin groups and the positive control group showed significantly increased E2,FSH,uterine endometrial thickness,and the number of uterine endometrial glands,significantly upregulated Wnt3a and β-catenin mRNA and protein ex-pression,reduced T,LH,TNF-α,IL-1β,and IL-6 levels,and markedly downregulated ER and PR protein expression(P<0.05).Conclu-sion:Paeoniflorin can regulate hormone levels in PCOS rats,reduce inflammatory responses,improve uterine endometrial receptivity,and its effects may be achieved by influencing the Wnt/β-catenin pathway.
