毛冬青片对脑缺血再灌注损伤大鼠的作用及对GFAP、NF-κBp65、VEGFA蛋白表达的影响

目的:观察毛冬青片对脑缺血再灌注损伤大鼠的治疗作用及对GFAP、NF-κB p65、VEGFA蛋白表达的影响,探讨毛冬青片的促血管新生及神经保护作用。方法:线栓法复制MCAO/R大鼠模型,60只SD大鼠随机分为:正常对照组、模型对照组、银杏叶提取物20 mg/kg组、毛冬青片100、200 mg/kg组;造模后第24 h、3 d、7 d采用Longa评分和平衡木评分评估大鼠神经功能;HE染色观察组织病理学改变;免疫荧光染色法观察梗死灶周围脑组织CD31、VEGFA阳性表达情况;免疫荧光双染法观察GFAP/Ne-uN、NF-κB p65/NeuN 阳性共表达情况;Western blot 法检测梗死灶周围脑组织 GFAP、NF-κB p65、Phospho-NF-κB p65、VEGF-R2 蛋白表达。结果:与正常对照组比较,模型对照组大鼠造模后各时间点Longa评分和平衡木评分显著升高(P<0。01);脑组织缺血核心区出现坏死性改变,神经元数目明显减少且排列紊乱,细胞呈空泡样改变;梗死灶周围脑组织NF-κB p65阳性细胞总数和核易位数、GFAP阳性细胞数、CD31和VEGFA阳性血管数明显增加(P<0。05或P<0。01),NeuN阳性细胞数显著减少(P<0。01);NF-κB p65、Phospho-NF-κB p65、GFAP、VEGF-R2蛋白表达明显上调(P<0。05或P<0。01);与模型对照组比较,毛冬青片200 mg/kg组能显著降低造模后第3、7 d的Longa评分和平衡木评分(P<0。01);明显减轻脑缺血再灌注大鼠的组织病理学损伤;明显降低NF-κB p65阳性细胞总数和核易位数、GFAP阳性细胞数和蛋白表达、Phospho-NF-κB p65蛋白表达(P<0。05或P<0。01),明显增加NeuN细胞数、CD31和VEGFA阳性血管数,明显上调VEGF-R2蛋白表达(P<0。05或P<0。01)。结论:毛冬青片通过下调GFAP蛋白表达抑制星形胶质细胞过度活化;下调NF-κB p65的核易位和磷酸化表达减轻炎症反应;上调梗死灶脑组织CD31、VEGFA、VEGF-R2蛋白表达,促进血管新生,从而减轻脑缺血再灌注损伤和组织病理学损伤,发挥神经保护作用。
Effect of Maodongqing(毛冬青)Tablets on Rats with Cerebral Ischemia-Reperfusion Injury and the Expressions of GFAP,NF-κBp65 and VEGFA Proteins
Objective:To observe the therapeutic effect of Maodongqing(毛冬青)Tablets on rats with cerebral ischemia-reperfusion injury and the expressions of GFAP,NF-κBp65 and VEGFA proteins,to explore the pro-angiogenic and neuroprotective effects of the prescription.Meth-ods:The MCAO/R rat model was replicated by suture method.Sixty SD rats were randomly divided into normal control group,model control group,Ginkgo biloba extract group,and Maodongqing Tablets groups(100 and 200 mg/kg).The neurological function of rats was evaluated by Longa score and balance beam score at 24 h,3 d and 7 d after modeling.The histopathological changes were detected by HE staining.Im-munofluorescence staining was used to observe the positive expressions of CD31 and VEGFA in the brain tissue around the infarct.The posi-tive co-expressions of GFAP/NeuN and NF-κBp65/NeuN were measured by immunofluorescence double staining.Western blot was performed to detect the expressions of GFAP,NF-KBp65,Phospho-NF-KBp65 and VEGF-R2 proteins in the brain tissue around the infarct.Results:Compared with the normal control group,rats in the model control group showed higher Longa scores and balance beam scores at all time points after modeling(P<0.01),with necrotic changes in the ischemic core of the brain tissue,a significant reduction in the number of neu-rons and disorganization,and vacuolated cells.The total number of NF-KBp65-positive cells,as well as the number of nuclear translocations in the brain tissue around the infarcted foci,GFAP-positive cells,CD31 and VEGFA-positive blood vessels were increased(P<0.05 or P<0.01),while the number of NeuN-positive cells was decreased(P<0.01).The expressions of NF-κBp65,Phospho-NF-κBp65,GFAP,and VEGF-R2 proteins were up-regulated(P<0.05 or P<0.01).Compared with the model control group,Maodongqing Tablets group(200 mg/kg)lowered the Longa score and balance beam score on the 3rd and 7th day after modeling(P<0.01),and reduced the histopatho-logical injury in cerebral ischemia-reperfusion rats.The total number of NF-κBp65-positive cells,the number of nuclear translocations,the number of GFAP-positive cells and protein expression,and the expression of Phospho-NF-κ Bp65 protein were decreased(P<0.05 or P<0.01),while the number of NeuN cells and CD31-and VEGFA-positive blood vessels,and the VEGF-R2 protein expression were increased(P<0.05 or P<0.01).Conclusion:Maodongqing Tablets can inhibit the over-activation of astrocytes by down-regulating the GFAP protein,attenuate the inflammatory response by down-regulating the nuclear translocation and phosphorylation of NF-κBp65,and promote angiogenesis by up-regulating CD31,VEGFA and VEGF-R2 proteins.Therefore,it relieves cerebral ischemia-reperfusion injury and histopathological dam-age,and plays a neuroprotective role.
