Protective Effect and Antioxidant Activity of Artemisia scoparia Extract on LPS-Induced Acute Lung Injury in Mice
Objective:To study the protective effect of Artemisia scoparia extract on lipopolysaccharide(LPS)-acute lung injury(ALI)in mice,and evaluate its antioxidant activity in vitro.Methods:Sixty ICR mice were randomly divided into normal control group,model control group,dexa-methasone acetate(2.5 mg/kg)group,and A.scoparia extract(100,200,400 mg/kg)groups.Drugs were continuously administered once a day for five days,and at 1.5 hours after the last administration,LPS(10 mg/kg)was dripped into the nasal cavity of mice to establish mouse ALI model.After 16 hours of modeling,lung index was measured,and myeloperoxidase(MPO),superoxide dismutase(SOD),glutathione(GSH)and malondialdehyde(MDA)were detected by the kits.Pathological changes of lung tissue in mice were observed by hematoxylin-e-osin(HE)staining.The scavenging assay of DPPH,ABTS and hydroxyl free radicals and ferric reducing ability assay were carried out to e-valuate the antioxidant activity of A.scoparia extract in vitro.Results:Compared with the normal control group,the model control group had decreased body weight and serum SOD activity(P<0.05)as well as elevated lung index and serum MPO,MDA and GSH(P<0.05 or P<0.01).Significant pathological changes were observed in lung tissue,including incrassation of alveolar wall,alveolar hemorrhage and inflam-matory cell infiltration.Compared with the model control group,A.scoparia extract 200 and 400 mg/kg down-regulated serum MPO and GSH(P<0.05),A.scoparia extract 400 mg/kg reduced the lung index and serum MDA(P<0.05),increased serum SOD(P<0.05),and allevi-ated lung injury in mice.In vitro antioxidant assay of A.scoparia extract,its IC50 of DPPH,ABTS and hydroxyl free radicals were 24.46,17.70 and 347.3 μg/mL,respectively.The results indicated that A.scoparia extract showed significant scavenging ability against free radi-cals in vitro in a dose-dependent manner in a certain range.Additionally,It has certain reducing ability.Conclusion:A.scoparia extract has antioxidant activity in vitro and exerts obvious protective effect on ALI mice,and the mechanism may be related to its excellent antioxidant effect.
