目的:观察蛭龙活血通瘀胶囊通过调控NLRP3信号途径对高血压心肌纤维化小鼠的影响及机制研究.方法:经皮下渗透压缓释泵持续性灌注外源性Ang Ⅱ 1.46 mg/kg 28 d制备高血压心肌纤维化小鼠模型,造模后随机分为:模型对照组、蛭龙活血通瘀胶囊1.56、3.12、6.24 g/kg组、福辛普利钠2.6×10-3 g/kg组,另设正常对照组.各组分别给予相应药物或生理盐水,1次/d,持续4 w,每周测量血压,根据动脉收缩压水平是否达150 mmHg以上和马松染色结果评估造模结果.第4 w后测量并分析左室射血分数(LVEF)、左室短轴缩短率(LVFS)、左心室收缩末期内径(LVESD)和左心室舒张末期内径(LVEDD)等心功能指标;酶联免疫吸附法(ELISA)检测血清中脑钠肽(BNP)和白细胞介素-1β(IL-1β)的含量;HE、Masson染色观察心肌细胞形态及胶原纤维的变化;免疫组织化学染色检测纤维连接蛋白(FN)、NOD样受体热蛋白结构域相关蛋白3(NLRP3)的阳性表达;蛋白质免疫印迹法(West-ern blot)和实时荧光定量聚合酶链反应(qRT-PCR)分别检测FN、α平滑肌肌动蛋白(α-SMA)、NLRP3、半胱氨酸天冬氨酸蛋白水解酶-1(Caspase-1)、IL-1β、凋亡相关斑点样蛋白(ASC)等mRNA和蛋白表达.结果:输注Ang Ⅱ后小鼠收缩压均达150 mmHg,Masson结果显示心肌出现大量蓝色胶原纤维,表明造模成功;与正常对照组比较,模型对照组小鼠的LVEF、LVFS指数显著减低,LVEDD、LVESD指数显著升高(P<0.01),心肌细胞肥大,排列紊乱,可见大量蓝色胶原纤维,BNP、IL-1β含量显著增高(P<0.01),F、Asma、Nlrp3、Caspasel、Il1b、Asc的mRNA和蛋白表达显著上调(P<0.01);与模型对照组比较,各药物组的LVEF、LVFS显著升高(P<0.01),LVEDD、LVESD指数明显降低(P<0.05),心肌细胞肥大及排列紊乱明显改善,心肌蓝色胶原纤维面积明显减少,BNP、IL-1β含量显著降低(P<0.01),Fn、Asma、Nlrp3、Caspase1、Il1b、Asc的mRNA和蛋白表达显著下调(P<0.01).结论:蛭龙活血通瘀胶囊可以改善高血压心肌纤维小鼠的心功能损伤,其作用机制可能与调控NLRP3信号通路,抑制NLRP3炎症小体的激活,进而减轻炎症因子的浸润有关.
Effect of Zhilong Huoxue Tongyu(蛭龙活血通瘀)Capsule on Hypertensive Myocardial Fibrosis by Regulating NLRP3 Signal Pathway
Objective:To study the effect and mechanism of the Zhilong Huoxue Tongyu(蛭龙活血通瘀胶囊)Capsule on mice with hypertensive myocardial fibrosis by regulating the NLRP3 signaling pathway.Methods:In this study,the mouse model of hypertensive myocardial fibrosis was established by continuous infusion of exogenous Ang Ⅱ of 1.46 mg/kg for 28 d through a subcutaneous osmotic sustained release pump.After modeling,the mice were randomly divided into a normal control group,a model control group,Zhilong Huoxue Tongyu Capsule(1.56 g/kg,3.12 g/kg,and 6.24 g/kg)groups,and fosinopril sodium(2.6×10-3 g/kg)group.Mice were given with corresponding drugs or normal saline,once a day for four weeks.At the same time,blood pressure was measured every week.The results of the model were evaluated accord-ing to whether the arterial systolic blood pressure level was above 150 mmHg and the results of Masson staining.After four weeks,left ventricu-lar ejection fraction(LVEF),left ventricular fraction shortening(LVFS),Left ventricular end-systolic dimension(LVESD),left ventricular end-diastolic dimension(LVEDD),and other cardiac function indexes were measured and analyzed.Enzyme-linked immunosorbent assay(ELISA)was used to detect serum levels of brain natriuretic peptide(BNP)and interleukin(IL)-1β.The changes in cardiomyocyte mor-phology and collagen fibers were observed by hematoxylin-eosin(HE)and Masson staining.Immunohistochemical staining was used to detect the positive expressions of fibronectin(FN)and NOD-like receptor thermal protein domain associated protein 3(NLRP3).Western blot and quantitative real-time polymerase chain reaction(qRT-PCR)were used to detect the protein and mRNA expressions of FN,α-smooth muscle actin(α-SMA),NLRP3,Cysteinyl aspartate specific proteinase-1(Caspase-1),IL-1β,and apoptosis-associated speck-like protein containing CARD(Asc).Results:After infusion of Ang Ⅱ,the systolic blood pressure of mice reached 150 mmHg.Masson results showed a large number of blue collagen fibers,indicating successful molding.Compared with the normal control group,the LVEF and LVFS indexes of the model con-trol group were decreased significantly,while the LVEDD and LVESD indexes were increased significantly(P<0.01).Cardiomyocytes are hy-pertrophic and disordered,with a large number of blue collagen fibers.The content of BNP and IL-1β was increased significantly(P<0 01).The mRNA and protein expressions of Fn,α-Sma,Nlrp3,Caspase-1,Il-1β,and Asc were significantly up-regulated(P<0.01).Compared with the model control group,the LVEF and LVFS in each drug group were increased significantly(P<0 01),while the LVEDD and LVESD inde-xes were decreased significantly(P<0.05).The hypertrophy and disorder of cardiomyocytes were significantly improved.The area of blue col-lagen fibers in the myocardium was decreased significantly.The content of BNP and IL-1β was decreased(P<0.01).The mRNA and protein expressions of Fn,α-Sma,Nlrp3,Caspase-1,IL-1β,and Asc were significantly down-regulated(P<0.01).Conclusion:Zhilong HuoxueTongyu Capsule can improve the cardiac function injury of mice with hypertensive myocardial fiber,and its mechanism may be related to regulating the NLRP3 signal pathway,inhibiting the activation of NLRP3 inflammasomes,and then reducing the infiltration of inflammatory factors.
