基于数据挖掘的结直肠癌动物模型应用特点分析

目的:研究结直肠癌动物模型的应用情况,为其动物实验方法和模型完善提供参考。方法:以结直肠癌和动物模型为主题词,在中国知网、万方数据库、PubMed中收集2001年至2021年结直肠癌动物模型的相关文献,总结实验动物种类、性别、造模方法、癌细胞种类、阳性药、检测指标等,建立数据库进行统计分析。结果:共筛选得到符合标准的文献274篇,其中实验动物多为4~6周龄、6~8周龄的BALB/c小鼠,以雄性居多;造模方法多采用移植法,在腋下接种最为常见;药物诱导造模法中多以AOM/DSS为诱导剂造模。造模癌细胞以鼠源的CT-26细胞和人源的Lovo细胞使用较多,阳性药多选择5-氟尿嘧啶,剂量以20 mg/kg为主,给药周期多为4 w;高频检测指标主要包括肿瘤组织表观指标、小鼠表观指标、肿瘤组织病理、肿瘤组织免疫组化、肿瘤组织蛋白免疫印迹法、抑瘤率。结论:目前结直肠癌动物模型造模方法单一,建议增加中医因素干预,构建造模成功率高、重现性好、与临床吻合度高的动物模型,本研究通过对结直肠癌动物试验进行挖掘分析,对不同动物模型进行评估,通过挖掘内容为模型应用提供参考,为模型完善提供思路。
Application of Colorectal Cancer Animal Model:Based on Data Mining
Objective:To study the application of animal models of colorectal cancer and provide a reference for improving the animal experiment method and animal model.Methods:With colorectal cancer and animal model as subject terms,we retrieved articles on animal models of colorectal cancer from China National Knowledge Infrastructure(CNKI),Wanfang Data,and PubMed(2001-2021)summarized the types and genders of experimental animals,modeling methods,cancer cell types,positive drugs,and detection indexes,and established a database for sta-tistical analysis.Results:A total of 274 eligible papers were screened out.The majority of the involved experimental animals were male BALB/c mice of 4-6 weeks old or(6-8)weeks old.The most frequently used modeling method was transplantation(generally by axillary in-oculation).As for the modeling with drugs,AOM/DSS were favored.Mouse CT-26 cell line and human LoVo cell line were common cells for the experiments.The 5-fluorouracil was frequently selected as the positive drug,generally with the dosage of 20 mg/kg and the administration usually lasted 4 weeks.The indexes of tumor tissue apparent index,mouse apparent index,tumor histopathology,immunohistochemistry result of tumor tissue,expression of tumor tissue proteins(Western blotting),and tumor inhibition rate were frequently adopted.Conclusion:A few methods for the animal modeling of colorectal cancer are available.It is suggested that traditional Chinese medicine should be involved in the modeling so as to improve the success rate of modeling,reproducibility of models,and similarity to the disease in clinical settings.This study evaluates different animal models of colorectal cancer based on data mining,which is expected to serve as a reference for model application and model improvement.
