Anti-Inflammatory Effect of the Extract of Fresh Periplaneta americana
Objective:To study the anti-inflammatory effect of the extract of fresh Periplaneta americana.Methods;Different models of inflamma-tion were treated with the extract of fresh P.americana,and the effects of the extract on relevant indicators were investigated.The RAW264.7 cell model of inflammation was established with lipopolysaccharide(LPS),and the release of interleukin-6(IL-6)and nitric oxide(NO)was measured.The blood vessel permeability,ear swelling,and foot swelling were measured in the animal models of acute inflammation estab-lished with xylene,croton oil,and carrageenan,respectively.A mouse model of chronic inflammation was established by inoculation of cotton balls into the armpit,and the dry weight of granuloma was measured.The levels of interleukin-6(IL-6)and prostaglandin E2(PGE2)in the serum of the rat model of toe swelling induced by carrageenan were determined by enzyme-linked immunosorbent assay.Results:The extract of fresh P.americana inhibited the LPS-induced release of IL-6 and NO in RAW264.7 cells(P＜0.01).It curbed the xylene-induced in-crease in skin capillary permeability in mice(P＜0.01),relieved the ear swelling induced by croton oil in mice(P＜0.01),reduced the dry weight of granuloma in the mouse model of chronic inflammation(P＜0.01),and alleviated the toe swelling induced by carrageenan in rats(P＜0.05 or P＜0.01).Furthermore,the extract lowered the serum level of IL-6 in rats(P＜0.05).Conclusion:The extract of fresh P.ameri-cana demonstrated significant anti-inflammatory effect.
