美洲大蠊鲜虫提取物的抗炎作用研究

目的:研究美洲大蠊鲜虫提取物的抗炎作用。方法:用美洲大蠊鲜虫提取物处理不同炎症模型,检测其对相关指标的影响。用脂多糖(LPS)刺激小鼠巨噬细胞RAW264。7造体外炎症模型,检测一氧化氮(NO)、白细胞介素6(IL-6)的释放。用二甲苯、巴豆油、角叉菜胶造动物急性炎症模型,分别检测血管通透性、耳廓肿胀度和足肿胀度;用棉球接种于小鼠腋下造慢性炎症模型,称肉芽干质量。角叉菜胶足趾肿胀试验中用酶联免疫法检测大鼠血清中IL-6和前列腺素E2(PGE2)的含量。结果:美洲大蠊鲜虫提取物可显著抑制LPS诱导的RAW264。7细胞释放NO和IL-6(P<0。01);抑制二甲苯致小鼠皮肤毛细血管通透性增高(P<0。01);减轻巴豆油致小鼠耳廓肿胀度(P<0。01)、减轻慢性炎症模型小鼠的肉芽肿干质量(P<0。01)、减轻角叉菜胶致大鼠足肿胀度(P<0。05或P<0。01),显著降低大鼠血清中IL-6含量(P<0。05)。结论:美洲大蠊鲜虫提取物具有明显的抗炎作用。
Anti-Inflammatory Effect of the Extract of Fresh Periplaneta americana
Objective:To study the anti-inflammatory effect of the extract of fresh Periplaneta americana.Methods;Different models of inflamma-tion were treated with the extract of fresh P.americana,and the effects of the extract on relevant indicators were investigated.The RAW264.7 cell model of inflammation was established with lipopolysaccharide(LPS),and the release of interleukin-6(IL-6)and nitric oxide(NO)was measured.The blood vessel permeability,ear swelling,and foot swelling were measured in the animal models of acute inflammation estab-lished with xylene,croton oil,and carrageenan,respectively.A mouse model of chronic inflammation was established by inoculation of cotton balls into the armpit,and the dry weight of granuloma was measured.The levels of interleukin-6(IL-6)and prostaglandin E2(PGE2)in the serum of the rat model of toe swelling induced by carrageenan were determined by enzyme-linked immunosorbent assay.Results:The extract of fresh P.americana inhibited the LPS-induced release of IL-6 and NO in RAW264.7 cells(P<0.01).It curbed the xylene-induced in-crease in skin capillary permeability in mice(P<0.01),relieved the ear swelling induced by croton oil in mice(P<0.01),reduced the dry weight of granuloma in the mouse model of chronic inflammation(P<0.01),and alleviated the toe swelling induced by carrageenan in rats(P<0.05 or P<0.01).Furthermore,the extract lowered the serum level of IL-6 in rats(P<0.05).Conclusion:The extract of fresh P.ameri-cana demonstrated significant anti-inflammatory effect.
