Establishment of Fingerprints and Spectrum-Effect(Relieving Migraine)Relationship of Baizhi(白芷)Produced in Different Regions
Objective:To establish the HPLC fingerprints of Baizhi(白芷)and explore the pharmacodynamic material basis for the migraine-relie-ving effect of Baizhi based on the spectrum-effect relationship.Methods:The fingerprints of 11 batches of Baizhi samples were established by HPLC,and the SOP of Similarity evaluation system for chromatographic fingerprint of TCM(version 2012)was used to evaluate the similarity of the fingerprints.The rat model of nitroglycerin-induced migraine was used to examine the effects of Baizhi on rat behaviors,and the content of 5-hydroxytryptamine(5-HT),calcitonin gene-related peptide(CGRP),and nitric oxide(NO)in the plasma and dopamine(DA)and nor-epinephrine(NE)in the brain tissue was determined by ELISA.The pharmacodynamic material basis of Baizhi for relieving migraine was comprehensively analyzed by principal component analysis,gray correlation analysis,and the entropy weight method.Results:A total of 17 common peaks were identified in the fingerprints of 11 batches of Baizhi samples,among which 9 components were identified by comparison with reference substances.Baizhi reduced the number of head scratches,lowered the levels of 5-HT,CGRP,NO and NE,and elevated the lev-el of DA in the rat model of migraine.The spectrum-effect correlation analysis showcased that isoimperatorin,byakangelicin,imperatorin,and phellopterin contributed the most to the migraine-relieving effect,which may be the key pharmacodynamic material basis for the migraine-relie-ving effect of Baizhi.Conclusion:The HPLC fingerprints of Baizhi were established,and the pharmacodynamic material basis of Baizhi for re-lieving migraine was initially clarified based on the spectrum-effect relationship.The findings can provide a reference for the research on the pharmacodynamic material basis and the quality evaluation of Baizhi.
