温胆汤对精神分裂症模型大鼠肠道菌群失调的干预作用

目的:探讨温胆汤对菌群失调的精神分裂症模型大鼠肠道菌群、短链脂肪酸(SCFA)及额叶皮质中 γ-氨基丁酸(GA-BA)、五羟色胺(5-HT)、多巴胺(DA)含量的影响。方法:将70只大鼠随机分为正常对照组、MK-801组、MK-801+抗生素互作模型组、MK-801+抗生素+温胆汤(40、20、10 g/kg)组和MK-801+抗生素+氯氮平0。02 g/kg组,每组10只。温胆汤10、20、40 g/kg、氯氮平0。02 g/kg分别灌胃相应药物,连续21 d,抗生素灌胃给予氨苄青霉素200 mg/kg、硫酸新霉素200 mg/kg、甲硝唑200 mg/kg、盐酸万古霉素100 mg/kg混悬液,连续28 d,正常对照组和MK-801组灌胃等体积的生理盐水。末次给药2 h后,除正常对照组外,其余组大鼠腹腔注射0。6 mg/kg MK-801,建立精神分裂症模型。观察大鼠刻板行为,采用16S rDNA测序分析菌群变化,GC-MS/MS法检测短链脂肪酸的含量,LC-MS/MS法测定额叶皮质中神经递质含量。结果:与正常对照组相比,30 min后MK-801+抗生素互作模型组刻板行为评分显著上升,肠道菌群数量、多样性及差异物种下降,盲肠内容物中乙酸、丙酸、丁酸、异丁酸、异戊酸含量显著下降(P<0。01),脑组织中GABA、5-HT、DA含量显著下降(P<0。01);与MK-801+抗生素互作模型组相比,各给药组刻板行为评分明显降低(P<0。05),温胆汤治疗后大鼠肠道菌群数量、多样性及差异物种增加;乙酸、异丁酸、异戊酸含量增加(P<0。05);MK-801+抗生素+温胆汤10、40 g/kg组脑组织中GABA、5-HT、DA含量明显升高,MK-801+抗生素+温胆汤20 g/kg组5-HT含量明显升高(P<0。05)。结论:温胆汤可能通过调节MK-801+抗生素互作模型大鼠的刻板行为、改善肠道菌群失调、增加短链脂肪酸及脑组织GABA、5-HT、DA含量,从而达到改善菌群失调的精神分裂症模型大鼠的学习认知能力。
Wendan (温胆) Decoction Regulates Intestinal Flora Disturbance in Rat Model of Schizophrenia
Objective:To investigate the effects of Wendan (温胆) Decoction on the intestinal flora and the expression of short-chain fatty acids (SCFAs),gamma-aminobutyric acid (GABA),5-hydroxytryptamine (5-HT),and dopamine (DA) in the rat model of schizophrenia with in-testinal flora disturbance.Methods:Seventy rats were randomized into normal control,MK-801,MK-801+antibiotics,MK-801+antibiotics+Wendan Decoction (40,20,and 10 g/kg),and MK-801+antibiotics+clozapine (0.02 g/kg) groups,with 10 rats in each group.The rats in drug intervention groups were administrated corresponding drugs by gavage for 21 days,and the antibiotic suspension composed of ampicillin (200 mg/kg),neomycin sulfate (200 mg/kg),metronidazole (200 mg/kg),vancomycin hydrochloride (100 mg/kg) was administrated by gavage for 28 days.The rats in the normal control group and MK-801 group were administrated with the same volume of normal saline.The rats in other groups except the normal control group were modeled for schizophrenia by intraperitoneal injection with 0.6 mg/kg MK-8012 h after the last administration.The stereotyped behavior of rats was observed.The 16S rDNA sequencing was employed to analyze the composi-tion of intestinal flora.The content of SCFAs and neurotransmitters in the frontal cortex was measured by GC-MS/MS and LC-MS/MS,respec-tively.Results:Compared with the normal control group,the MK-801+antibiotics group showed increased stereotyping behavior score after 30 min,decreased number,diversity,and differential species of intestinal flora,reduced content of acetic acid,propionic acid,butyric acid,isobutyric acid,and isovaleric acid (P<0.01),and lowered levels of GABA,5-HT,and DA (P<0.01).Compared with the MK-801+antibi-otics group,drug interventions decreased the stereotyping behavior score (P<0.05).The treatment with Wendan Decoction increased the number,diversity and differential species of intestinal flora and the content of acetic acid,isobutyric acid,and isovaleric acid (P<0.05).The MK-801+antibiotics+Wendan Decoction (40 and 10 g/kg) groups showed increased content of GABA,5-HT,and DA,while the MK-801+antibiotics+Wendan Decoction (20 g/kg) group only showed increased 5-HT content (P<0.05).Conclusion:Wendan Decoction may im-prove the learning and cognitive ability in the rat model of schizophrenia with intestinal flora disturbance by regulating the stereotyped behav-ior,repairing the intestinal flora disturbance,and increasing the content of SCFAs and GABA,5-HT and DA in the brain tissue of the rats trea-ted with MK-801 and antibiotics.
