Mechanism of Modified Suanmei Decoction in the Treatment of MAFLD Based on Transcriptomics and Metabolomics
Objective:To investigate the therapeutic effect and mechanism of modified Suanmei Decoction on metabolic-associated fatty liver disease (MAFLD).Methods:C57BL6 J mice were randomly divided into normal mice and model mice.The model mice were fed a high-fat diet for 12 weeks to induce MAFLD.After successful modeling,the model mice were randomly divided into model control group,modified Suanmei Decoction groups (0.55,1.1,and 2.2 g/kg),and metformin group (0.10 g/kg),with 6 mice in each group.Changes in body weight be-fore and after treatment,liver weight,and liver index after treatment were observed.Biochemical methods were used to detect serum total cholesterol (TC),triglycerides (TG),low-density lipoprotein (LDL-C),high-density lipoprotein (HDL-C),and liver TC and TG levels.HE and oil red O staining were used to observe liver pathology and lipid deposition.Transcriptomics and metabolomics were employed to ex-amine changes in gene expression and small molecule metabolites in the liver.RT-qPCR was used to detect the expression of Pi3k and Akt mRNA in the liver.Results:Compared with the normal control group,the model control group showed an increase in body weight and liver index,significantly higher levels of serum TC,TG,LDL-C,and liver TC and TG,and decreased serum HDL-C.Fatty degeneration and nu-merous lipid droplets were observed in the liver,and Pi3k and Akt mRNA expression was significantly upregulated (P<0.05).Compared with the model control group,the modified Suanmei Decoction groups showed reduced body weight,liver weight,liver index,and blood lipid levels.Liver pathology indicated a reduction in lipid deposition and fewer lipid droplets in the liver tissue.Transcriptomic KEGG enrichment analysis suggested that the improvement of MAFLD by modified Suanmei Decoction may be related to pathways involving lipid metabolism and inflammation,with a significant decrease in Pi3k and Akt mRNA expression in the liver (P<0.05).Conclusion:Modified Suanmei Decoc-tion can improve MAFLD in mice,and its mechanism may be related to the inhibition of the PI3K-AKT pathway.
