欢草汤对乳腺增生症模型大鼠性激素及其受体表达的影响

目的:探索欢草汤对乳腺增生症模型大鼠的治疗作用和机制。方法:80只大鼠随机选取10只作为正常对照组,其余大鼠造成乳腺增生症模型。将造模成功的大鼠随机分为模型对照组、欢草汤6。1、12。1、24。2 g/kg组、欢草汤12。1 g/kg治未病组和他莫昔芬4 mg/kg组。连续治疗4 w后,观察各组大鼠乳房的外观并测量乳头的高度和直径;HE染色后观察乳腺组织病理形态;ELISA法测血清促性腺激素释放激素(GnRH)、卵泡刺激素(FSH)、黄体生成素(LH)、催乳素(PRL)、雌二醇(E2)、孕酮(P)和睾酮(T)的含量;Western blot法检测乳腺组织雌激素受体α(ERα)、雌激素受体β(ERβ)、孕激素受体(PR)、催乳素受体(PRLR)蛋白的表达;RT-qPCR法检测乳腺组织雌激素受体α(Esr1)、雌激素受体β(Esr2)、孕激素受体(Pgr)、孕激素受体A(Pgra)、孕激素受体B(Pgrb)和催乳素受体(Prlr)mRNA的表达。结果:与正常对照组比较,模型对照组大鼠血清E2和PRL含量显著升高,P含量显著降低(P<0。01);乳腺组织ERα、PR和PRLR蛋白表达显著上调,ERβ蛋白表达显著下调(P<0。01);乳腺组织Esr1、Pgr、Pgrb和Prlr mRNA表达显著上调,Esr2、Pgra mRNA表达明显下调(P<0。05或P<0。01)。予欢草汤治疗后,上述指标明显逆转(P<0。05或P<0。01)。结论:欢草汤可能通过降低血清E2和PRL及升高P水平,抑制乳腺组织ERα、PR、PRLR及促进ERβ蛋白表达,下调乳腺组织Esr1、Pgr、Pgrb和Prlr及促进Esr2和Pgra mRNA表达,治疗乳腺增生症。
Effect of Traditional Chinese Medicine Master Ban Xiuwen's Huancao Decoction (欢草汤) on Sex Hormone and Expression of Sex Hormone Receptor in Rats with Hyperplasia of Mammary Glands
Objective:To explore the therapeutic effect and therapeutic mechanism of Huancao Decoction (欢草汤) on rats with hyperplasia of mammary glands.Methods:A total of 10 of the 80 rats were randomly selected as the normal control group,while the rest rats were made into the model of hyperplasia of mammary glands.The successfully modeled rats were randomly divided into model control group,Huancao Decoc-tion (24.2,12.1,and 6.1 g/kg) groups,Huancao Decoction (12.1 g/kg) preventive treatment group,and Tamoxifen (4 mg/kg) group.Af-ter four weeks of continuous treatment,the appearance of the breasts of rats in each group was observed,and the height and diameter of the nipples were measured.Pathological morphology of breast tissue was observed by hematoxylin-eosin (HE) staining.The levels of serum gon-adotropin-releasing hormone (GnRH),follicle-stimulating hormone (FSH),luteinizing hormone (LH),prolactin (PRL),estradiol (E2),pro-gesterone (P),and testosterone (T) were measured by enzyme-linked immunosorbent assay (ELISA).The protein expressions of estrogen receptor alpha (ERα),estrogen receptor beta (ERβ),progesterone receptor (PR),and prolactin receptor (PRLR) in breast tissue were measured by Western Blot (WB).The mRNA expressions of estrogen receptor alpha (Esr1),estrogen receptor beta (Esr2),progesterone (Pgr),progesterone receptor A (Pgra),progesterone receptor B (Pgrb),and prolactin receptor (Prlr) in breast tissue were detected by RT-qPCR.Results:Compared with the normal control group,the levels of serum E2 and PRL in the model control group were significantly in-creased,while the level of P was significantly reduced (P<0.01).In breast tissue,the protein expressions of ERα,PR,and PRLR were up-regulated significantly,while ERβ was downregulated significantly (P<0.01).The mRNA expressions of Esr1,Pgr,Pgrb,and Prlr in breast tissue were upregulated significantly,while those of Esr2 and Pgra were significantly downregulated (P<0.01 or P<0.05).After treatment with Huancao Decoction,the above indexes were significantly reversed (P<0.01 or P<0.05).Conclusions:Huancao Decoction may treat hy-perplasia of mammary glands by reducing serum E2 and PRL,increasing P level,inhibiting ERα,PR,and PRLR,promoting ERβ protein ex-pression in breast tissue,inhibiting Esr1,Pgr,Pgrb,and Prlr,and improving Esr2 and Pgra mRNA expressions in breast tissue.
