乌梅对脾虚泄泻模型大鼠的作用研究

目的:探究乌梅对脾虚泄泻模型大鼠的止泻作用及其作用机制。方法:灌胃大黄水煎液复制脾虚泄泻大鼠模型,给予乌梅水煎液或蒙脱石散干预5 d,观察大鼠一般情况,评判大鼠布里斯托(Bristol)粪便性状评分、测定大鼠稀便率、大便含水率、小肠推进率、胃残留率;测定血清胃动素(MLT)、胃泌素(GAS)和结肠中Na+-K+-ATP酶含量;观察结肠病理切片;免疫荧光法检测结肠水通道蛋白3(AQP3)、AQP4的表达。结果:与正常对照组比较,模型对照组大鼠稀溏明显,Bristol粪便性状评分、稀便率和大便含水率均显著升高,体质量显著降低,小肠推进率显著升高,胃残留率显著降低,血清MTL含量明显升高,血清GAS含量显著降低,结肠Na+-K+-ATP酶含量显著降低(P<0。01),光镜下可见结肠组织明显的炎症浸润改变、黏膜缺损与腺体结构分离,结肠AQP3、AQP4蛋白相对表达量显著降低(P<0。01);与模型对照组比较,乌梅各剂量组大鼠粪便稀溏好转,Bristol粪便性状评分、稀便率和大便含水率显著降低,小肠推进率显著降低,胃残留率显著增加(P<0。01),血清MTL、GAS含量明显降低(P<0。05或P<0。01),结肠Na+-K+-ATP酶含量、结肠AQP4蛋白表达显著升高(P<0。01),光镜下可见结肠组织增生及黏膜损伤改善;乌梅水煎液10 g/kg组体质量明显升高(P<0。05),乌梅水煎液2。5、5 g/kg组结肠AQP3蛋白表达明显升高(P<0。05或P<0。01)。结论:乌梅对脾虚泄泻大鼠有显著止泻作用,其具体机制与降低MLT、GAS含量,抑制胃肠运动亢进;调节Na+-K+-ATP酶活性,增强AQP3、AQP4表达,降低大便含水率;改善粘膜损伤,改善炎症浸润表现等有关。
Effect of MUME FRUCTUS on Rat Model of Pixuxiexie (脾虚泄泻) Syndrome
Objective:To investigate the anti-diarrheal effect and mechanism of MUME FRUCTUS on the Pixuxiexie syndrome (PXXX) model rats.Methods:The PXXX rat model was established by gavage with RHEI RADIX ET RHIZOMA decoction.MUME FRUCTUS decoction or mont-morillonite powder was given for 5 days.The general conditions of the rats were observed,and Bristol stool form scale,fecal consistency,fecal water content,small intestinal propulsion rate,and gastric residue rate were measured.Serum motilin (MLT),gastrin (GAS),and colonic Na+-K+-ATPase content were measured.Colonic pathological sections were observed,and immunofluorescence was used to detect the expres-sion of colonic aquaporin Protein-3 (AQP3) and AQP4.Results:Compared with the normal control group,the model control group showed significant loose stools,with significantly increased Bristol stool form scale,fecal consistency,and fecal water content,while body weight signif-icantly decreased,small intestinal propulsion rate significantly increased,and gastric residue rate significantly decreased (P<0.01).Serum MLT content increased significantly (P<0.05),serum GAS content decreased significantly (P<0.01),colonic Na+-K+-ATPase content de-creased significantly (P<0.01),obvious inflammation infiltration changes,mucosal defects,and glandular structure separation were observed under light microscope,and the relative expression levels of colonic AQP3 and AQP4 proteins decreased significantly (P<0.01).Compared with the model control group,each dose group of MUME FRUCTUS showed improvement diarrhea in PXXX model,with significantly decreased Bristol stool form scale,fecal consistency,and fecal water content (P<0.01),significantly decreased small intestinal propulsion rate,signifi-cantly increased gastric residue rate (P<0.01),significantly decreased serum MLT and GAS content (P<0.05 or P<0.01),significantly in-creased colonic Na+-K+-ATPase content,significantly increased colonic AQP4 protein expression (P<0.01),proliferation of colonic tissue and improvement of mucosal damage were observed under light microscope.The body weight of rats in the MUME FRUCTUS decoction 10 g/kg group increased significantly (P<0.05),and the expression of colonic AQP3 in the MUME FRUCTUS decoction 2.5 g/kg and 5 g/kg groups increased significantly (P<0.05 or P<0.01).Conclusion:MUME FRUCTUS has a significant anti-diarrheal effect on PXXX rats.Its specific mechanism is related to the reduction of MLT,GAS content,inhibition of gastrointestinal hyperactivity,regulation of Na+-K+-ATPase activity,enhancement of AQP3,AQP4 expression,and reduction of fecal water content.It also improves mucosal damage,inflamma-tion infiltration,and other manifestations.
