Spectrum-Effect Relationship of Danggui Buxue Decoction (当归补血汤) for Hematopoietic Effect
Objective:To investigate the correlation between the HPLC fingerprints and the hematopoietic effects of different extracts from Danggui Buxue Decoction(当归补血汤),to identify the active components responsible for the effects,and to determine the optimal extraction method.Methods:HPLC was utilized to establish the fingerprints of various extracts from Danggui Buxue Decoction.A rat model of blood deficiency was replicated using subcutaneous injections of acetylphenylhydrazine (APH).The impact of the four extracts on the blood routine and serum hematopoietic factors in rats with blood deficiency was observed.Orthogonal partial least squares (OPLS) was applied to correlate common peak areas in the fingerprints with the data on comprehensive pharmacological efficacy.Results:The supernatants of water extract-alcohol pre-cipitation of Danggui Buxue Decoction exhibited the optimal hematopoietic effect (P<0.01),and twenty common peaks were identified in the HPLC fingerprints of the four extracts.Nine peaks showed a high positive correlation with the comprehensive efficacy,and their contributions to the hematopoietic effect,in a descending order,were peak 14 (ononin),peak 11 (calycosin-7-O-β-D-glucoside),peak 13 (senkyunolide H),peak 10 (ferulic acid),peak 8 (unknown),peak 6 (unknown),peak 9 (chlorogenic acid),peak 12 (senkyunolide I),and peak 18 (methylnissolin).Conclusion:The spectrum-effect relationship and the HPLC fingerprint of Danggui Buxue Decoction for its hematopoietic effect were established,identifying the main active components for hematopoietic effect.This provided a reference for further pharmacodynamic substance research and quality evaluation of Danggui Buxue Decoction.
