Comparative Analysis of Nucleoside Components and Antioxidant Activities of Their Extracts in Five Different Species of Sanghuangporus
Objective:To optimize the extraction process conditions of nucleoside components from Sanghuangporus,a high-performance liquid chro-matography-diode array detector (HPLC-DAD) method was established to determine the content and evaluate the in vitro antioxidant activity of cytidine,uridine,guanosine,and adenosine in five species of Sanghuangporus.Methods:The ultrasonic-assisted extraction process of nu-cleoside components was optimized using an orthogonal experimental design.The HPLC-DAD method was applied to quantitatively and quali-tatively analyze cytidine,uridine,guanosine,and adenosine in the extracts.In addition,the content of other components including total phenols and total polysaccharides in the extracts were determined using ultraviolet-visible spectrophotometry.Finally,the in vitro antioxidant activities of the extracts were evaluated through DPPH radical,ABTS cationic radical,and·OH radical scavenging assay.Results:The optimum ex-traction conditions for nucleoside components from Sanghuangporus were as follows:aqueous extraction,ultrasound time of 30 minutes,solid-liquid ratio of 1:50,and extraction temperature of 40 °C.The total nucleoside content of Sanghuangporus vaninii,Inonotus hispidus,Sang-huangporus baumii,Sanghuangporus lonicericola,and Sanghuangporus toxicodendri was 661.07 μg/g,11098.14 μg/g,415.23 μg/g,1047.84 μg/g,and 216.89 μg/g,respectively.There were also certain differences in the content of total phenols and total polysaccharides in the extracts of five species of Sanghuangporus.The extracts of five species of Sanghuangporus were capable of scavenging DPPH free radi-cals,ABTS cationic free radicals,and OH free radicals,with the best scavenging capacity against ABTS cationic free radicals.Among the five species of Sanghuangporus,Inonotus hispidus had the best antioxidant activity,followed by Sanghuangporus toxicodendri,Sanghuangporus vani-nii,and Sanghuangporus baumii.Sanghuangporus lonicericola had poor antioxidant activity.Conclusion:This study optimized the extraction process of nucleoside components by orthogonal experimental design,and the established detection method was stable and reliable.This study also systematically evaluated the in vitro antioxidant activities of the five species of Sanghuangporus,providing a theoretical basis for further development and utilization of Sanghuangporus.
