山茱萸山药水提物对环磷酰胺诱导的免疫抑制小鼠免疫功能及其肠道微生物的影响

目的:探究山茱萸山药水提物对环磷酰胺诱导的免疫抑制小鼠免疫功能及其肠道微生物的影响。方法:将小鼠随机分为正常对照组、模型对照组、左旋咪唑片0。08 g/kg组、山茱萸山药水提物0。43、0。86、1。73 g/kg组,除正常对照组外,造模小鼠于第1~3 d腹腔注射40 mg/kg环磷酰胺。第4 d灌胃相应药物,连续14 d。末次给药24 h后,检测小鼠脏器指数、脾脏NK细胞百分率、外周血T、B淋巴细胞亚群比例,分析各组小鼠肠道菌群变化。结果:与正常对照组比较,模型对照组小鼠的体质量、胸腺指数、脾脏指数、CD8+T、CD19、CD19/CD13明显降低,NK细胞百分率明显升高(P<0。05或P<0。01);与模型对照组相比,左旋咪唑片0。08 g/kg组NK淋巴细胞百分率显著降低(P<0。01),山茱萸山药水提物1。73 g/kg组外周血CD19/CD3的百分率明显升高(P<0。05)。与正常对照组相比,模型对照组肠道菌群α多样性Shannon明显降低(P<0。05),疣微菌门显著降低(P<0。01),厚壁菌门明显升高(P<0。05);与模型对照组比较,山茱萸山药水提物0。43 g/kg组肠道中疣微菌门显著升高(P<0。01),山茱萸山药水提物0。86 g/kg组厚壁菌门明显降低(P<0。05),左旋咪唑片0。08 g/kg组拟杆菌门显著升高(P<0。05);在属水平分类上,与模型对照组比较,左旋咪唑片0。08 g/kg组另枝菌属、杆菌属明显升高(P<0。05或P<0。01),山茱萸山药水提物0。86 g/kg组肠道中杆菌属明显升高、Lachnoclostridium 明显降低(P<0。05 或 P<0。01),山茱萸山药水提物 1。73 g/kg 组 Lachnoclostridium 显著降低(P<0。01)。结论:山茱萸山药水提物在一定程度上可以改善环磷酰胺诱导的小鼠免疫功能异常和肠道菌群失调。
Effects of CORNI FRUCTUS-DIOSCOREAE RHIZOMA Aqueous Extract on Immune Function and Intestinal Flora in Cyclophosphamide-Induced Immunosuppressed Mice
Objective:To investigate the effects of CORNI FRUCTUS-DIOSCOREAE RHIZOMA aqueous extract on the immune function and intesti-nal flora in cyclophosphamide-induced immunosuppressed mice.Methods:The mice were randomly divided into a normal control group,a model group,a 0.08 g/kg levamisole tablets group,and 0.43,0.86,and 1.73 g/kg CORNI FRUCTUS-DIOSCOREAE RHIZOMA aqueous ex-tract groups,with 10-11 mice in each group.Except for the normal control group,mice in the other groups were intraperitoneally injected with 40 mg/kg cyclophosphamide on the first three days.From the 4th day,the corresponding drugs were given by gavage for 14 consecutive days.After 24 hours of the last administration,the organ indexes,the percentage of NK cells in the spleen,the proportions of T and B lymphocyte subsets in peripheral blood of mice in each group were determined,and the changes in intestinal flora in mice of each group were analyzed.Results:Compared with those of the normal control group,the body weight,thymus index,spleen index,CD8+T,CD19,and CD 19/CD 13 were significantly decreased in the model control group,while the percentage of NK cells was significantly increased(P<0.05 or P<0.01).Com-pared with those of the model control group,NK lymphocytes in the levamisole tablets group were significantly decreased(P<0.01),while the CD19/CD13 ratio was significantly increased after the intervention by 1.73 g/kg CORNI FRUCTUS-DIOSCOREAE RHIZOMA aqueous ex-tract(P<0.05).Compared with the normal control group,the model control group showed a significantly decreased Shannon index(P<0.05),which reflected the alpha diversity of intestinal flora,significantly decreased Verrucomicrobia(P<0.01),and significantly increased Firmicutes(P<0.05).In comparison with the model control group,the 0.43 g/kg CORNI FRUCTUS-DIOSCOREAE RHIZOMA aqueous ex-tract group saw significantly increased Verrucomicrobia(P<0.01),the 0.86 g/kg CORNI FRUCTUS-DIOSCOREAE RHIZOMA aqueous ex-tract group witnessed significantly decreased Firmicutes(P<0.05),the 0.08 g/kg levamisole tablets group showed significantly increased Bacteroidetes(P<0.05).In terms of genus classification,compared with those of the model control group,Odoribacter and Alistipes in the 0.08 g/kg levamisole tablets group were significantly increased(P<0.05,P<0.01);Odoribacter was significantly increased(P<0.05),while Lachnoclostridium was significantly decreased(P<0.01)in the 0.86 g/kg CORNI FRUCTUS-DIOSCOREAE RHIZOMA aqueous ex-tract group,Lachnoclostridium was significantly decreased in the 1.73 g/kg CORNI FRUCTUS-DIOSCOREAE RHIZOMA aqueous extract group(P<0.01).Conclusion:CORNI FRUCTUS-DIOSCOREAE RHIZOMA aqueous extract can improve the immune function and intestinal flora dysfunction induced by cyclophosphamide in mice.
