Chemical Constituents of Ethyl Acetate Extract of Suaeda salsa(L.)
Objective:To isolate and identify the chemical constituents of Suaeda salsa(L.)and evaluate their cardioprotective activity using a zebrafish model.Methods:The plant material was extracted with 85%ethanol under reflux.The extract was sequentially partitioned with petroleum ether,ethyl acetate,and n-butanol.The ethyl acetate fraction was further purified using silica gel columns,LH-20 gel columns,and C18 preparative columns.The chemical structures were elucidated using NMR and HRESIMS techniques.A zebrafish model of cardiac injury was established using verapamil hydrochloride to assess the effects of the compounds on pericardial edema and venous congestion areas.Results:Eleven compounds were isolated and identified as quercetin(1),luteolin(2),apigenin(3),N-trans-feruloyltyramine(4),methyl ferulate(5),trans-ferulic acid(6),p-hydroxycinnamic acid(7),vanillic acid(8),protocatechuic acid(9),protocatechualdehyde(10),and p-hydroxybenzoic acid(11).Compounds 6 and 8 at concentrations of 100 μg/mL and 50 μg/mL significantly ameliorated verapamil-induced cardiac injury.Conclusion:Compounds 3-5,7,and 10 were isolated from this plant for the first time.The cardioprotective mechanisms of compounds 6 and 8 warrant further investigation.
