摘要
目的:对盐地碱蓬(Suaeda salsa L.)的化学成分进行分离和鉴定,并基于斑马鱼模型评价其心脏保护活性.方法:盐地碱蓬经85%乙醇回流提取,浸膏依次经石油醚、乙酸乙酯和正丁醇萃取,再将乙酸乙酯部位依次经硅胶柱、LH-20型凝胶柱以及C18型制备柱等进行系统分离;采用NMR和HRESIMS等技术鉴定化学结构;采用盐酸维拉帕米构建斑马鱼心脏损伤模型,并评价化合物对斑马鱼心包水肿和静脉瘀血面积的影响.结果:共分离出11个化合物,分别鉴定为槲皮素(1)、木犀草素(2)、芹菜素(3)、N-反式-阿魏酰酪胺(4)、阿魏酸甲酯(5)、反式阿魏酸(6)、对羟基肉桂酸(7)、香草酸(8)、原儿茶酸(9)、原儿茶醛(10)、对羟基苯甲酸(11).100 μg/mL和50 μg/mL浓度的化合物6和8可显著改善盐酸维拉帕米诱导的心脏损伤.结论:化合物3~5、7和10为首次从该植物中分离得到;化合物6和8的心脏保护活性机制可进一步进行研究.
Abstract
Objective:To isolate and identify the chemical constituents of Suaeda salsa(L.)and evaluate their cardioprotective activity using a zebrafish model.Methods:The plant material was extracted with 85%ethanol under reflux.The extract was sequentially partitioned with petroleum ether,ethyl acetate,and n-butanol.The ethyl acetate fraction was further purified using silica gel columns,LH-20 gel columns,and C18 preparative columns.The chemical structures were elucidated using NMR and HRESIMS techniques.A zebrafish model of cardiac injury was established using verapamil hydrochloride to assess the effects of the compounds on pericardial edema and venous congestion areas.Results:Eleven compounds were isolated and identified as quercetin(1),luteolin(2),apigenin(3),N-trans-feruloyltyramine(4),methyl ferulate(5),trans-ferulic acid(6),p-hydroxycinnamic acid(7),vanillic acid(8),protocatechuic acid(9),protocatechualdehyde(10),and p-hydroxybenzoic acid(11).Compounds 6 and 8 at concentrations of 100 μg/mL and 50 μg/mL significantly ameliorated verapamil-induced cardiac injury.Conclusion:Compounds 3-5,7,and 10 were isolated from this plant for the first time.The cardioprotective mechanisms of compounds 6 and 8 warrant further investigation.
