干姜中姜酚类成分的提取工艺研究

目的:以6、8、10姜酚含量为指标,考察干姜中姜酚类成分的提取方法,确定其最佳提取工艺.方法:建立UPLC法同时测定6、8、10姜酚的含量方法;对6、8、10姜酚的提取溶剂、提取方法进行单因素考察,确定其最佳提取溶剂和提取方法;采用L9(34)正交试验设计,以6、8、10姜酚的总含量和浸膏得率为指标,考察乙醇浓度、乙醇倍数和渗滤速度3个因素对提取结果的影响.结果:UPLC法测定6、8、10姜酚的含量的最佳条件为:色谱柱:Acquity BEH C18(2.1 mm×100mm,1.7 μm);流动相:乙腈-0.1%醋酸水溶液,流速为0.30 mL·min-;梯度洗脱;检测波长:280 nm.6、8、10姜酚的最佳提取溶剂为80%乙醇,最佳提取方法为渗漉法;最佳渗漉工艺为:12倍量80%乙醇渗漉,渗漉速度为5 mL/(kg·min).结论:正交试验优选出的干姜中姜酚的提取方法,提取率较高,具备实际可行性.
Objective:To study the best extraction method of gingerols from dried Zingiber Officinale Rosc..Methods:The solvent and methods of extraction were studied to determine the optimal extraction methods by single factor test.And the concentration and volume of alcohol,velocity of percolation were investigated by L9(34)orthogonal experiment.The total content of 6,8,10 gingerol and extract rate were chosen as investigation indexes.Results:6,8,10 gingerol best extraction solvent was 80% ethanol and the best extraction method was the percolation method.The optimal percolation process was:12 times volume of 80% alcohol at the percolation rate of 5 mL/(kg · min).Conclusion:The optimized extraction method for Zingiber Officinale Rosc has high extraction rate,actual feasibility and is suitable for the modern production.
