摘要

目的:通过观察田麻益智颗粒含药血清对GLu损伤的胎鼠海马神经元的保护作用,探讨田麻益智颗粒对脑损伤神经元的保护机制.方法:取孕7 d Wistar大鼠的胎鼠海马,进行体外培养,并对培养的神经干细胞进行神经元鉴定.部分细胞开始贴壁,贴壁细胞呈圆形结果培养24 h后,大部分细胞已经贴壁生长:分化3d后神经元胞体增大,突起延长并出现分枝,形成稀疏的网络,7d胞体大,突起长,分枝连接成网,相互联系增多细胞成熟.MTT结果表明,田麻益智颗粒含药血清保护组与Glu损伤模型组相比细胞活性明显升高(P<0.05).平均值,田麻益智颗粒中剂量组对Glu损伤的神经元细胞保护作用与尼莫地平组的保护作用接近,优于正常组及模型组.说明田麻益智颗粒含药血清对Glu损伤海马神经元有保护作用.免疫荧光结果半定量分析结果标记CAMKII的神经元细胞中,统计分析结果表明正常组平均灰度值为148.6±9.88、保护组灰度值151.0±12.37低于Glu损伤组171.5士6.87(P <0.05),说明保护组中CAMKII的含量与正常组接近,并且较Glu损伤模型组少,中剂量组田麻益智颗粒对谷氨酸损伤神的海马神经元的有较好的保护作用.

Abstract

Through the observation of Tilia benefits-wisdom granule of drug-containing serum with Glu damage the protective effects of fetal rat hippocampal neurons,discusses Tilia benefits-wisdom granule of protective mechanism of brain injury neurons.Methods:Get fetal rat hippocampus of pregnant 7 d Wistar rat for in vitro culture and identification of cultivation of neural stem cells are neurons.Some cells begin to stick wall,adherent cells showing rounded result after cultivating 24 h,most cells have already adherent growth.Differentiation 3 d,increase neuronal cell body,protuberant prolong and branch to form the sparse network.7 d cell body larger,spike length,branch connected into a net,contact each other cells mature and more.Determined by MTT results showed significantly cell activity increased (P < 0.05) that Tilia benefits-wisdom granule drug-containing serum protection group compared with Glu injury model group.For the average,the protective effect in dose group of Tilia benefits-wisdom granule to Glu injuryand neurons cell is close to that of nim horizon group,is better than that of normal group and model group.That Tilia benefits-wisdom granule drug-containing serum has its protects on Glu damage hippocampal neurons.Immunofluorescence results mark CAMKII semi-quantitative analysis of neurons,the statistical analysis results show that the average grey value of 148.6 ± 9.88 in normal group; the protection group set of grey value of 151.0 ± 12.37 below Glu injury group 171.5 ±6.87 (P <0.05),suggesting to protect the content of CAMKII,close to the normal group,and relatively less than Glu injury model group,which discusses Tilia benefits-wisdom granule of protective mechanism of brain injury neurons.
