软坚散结胶囊含药血清诱导人肾透明细胞癌7860细胞凋亡

摘要
目的:研究软坚散结胶囊对人肾透明细胞癌7860细胞系抑制和诱导凋亡的作用.方法:分别采用CCK-8检测细胞的生长抑制率,流式细胞仪、激光共聚焦显微镜观察软坚散结合药血清处理7860细胞后细胞凋亡和细胞周期阻滞的发生.结果:软坚散结胶囊低、中、高剂量含药血清均能抑制7860细胞增殖(P＜0.05),其抑制率与作用时间及血清含药浓度相关.中、高剂量含药血清处理7860细胞48h后凋亡率显著高于对照组(P＜0.05),同时低、中、高剂量含药血清处理7860细胞48 h后,G0/G1期细胞明显较对照组增加(P＜0.01),提示软坚散结胶囊可促进细胞凋亡,并且将细胞生长周期阻滞于G0/G1期.激光共聚焦显微镜下可见经染色的凋亡细胞增多且细胞出现凋亡形态学改变.结论:软坚散结胶囊对人肾透明细胞癌7860细胞有抑制作用,机制可能与抑制细胞生长、阻滞细胞生长周期及诱导细胞凋亡有关.
Abstract
Objective:To study the growth inhibition and apoptosis effect of Ruanjian Sanjie Capsule on the renal clear cell cancer 7860 cells.Methods:CCK-8 assay was used to determine the cell growth inhibitory rate in vitro.Changes of apoptosis and cell cycle of 7860 cells after Ruanjian Sanjie Capsule treatment were observed by using flow cytometry and confocal laser scanning microscopy.Results:CCK-8 showed that serum containing Ruanjian Sanjie Capsule with low,middle and high doses inhibited the proliferation of 7860 cells (P ＜ 0.05) in a dose-and-time-dependent manner.In the middle and high dose treatment groups,7860 cells apoptosis rate after 48 h was significantly higher than those in the negative control group (P ＜ 0.05) while in low,middle and high dose treatment groups,G0/G1 phase after 48 h was obviously higher than that in the control group (P ＜ 0.01),indicating that the cell cycle was arrested in G0/G1 phase and promoting cells apoptosis.Through laser scanning confocal microscope,we observed the dyed apoptotic cells increased and morphological apoptosis was seen.Conclusion:Ruanjian Sanjie Capsule inhibited the proliferation of 7860 cells and its mechanism might be associated with inhibiting the cell growth,stagnation cell cycle and inducing apoptosis of 7860 cells.
