Effect of Traditional Chinese Medicine"Nourishing Yin and Subduing Yang"Method on PI3K/Akt Signal Pathway,Sexual Hormones and Insulin Related Indicators in Rats with Polycystic Ovarian Syndrome and Insulin Resistance
Objective To analyze the effect of traditional Chinese medicine nourishing Yin and subduing Yang method on the phosphoinositide 3-kinase/protein kinase B(PI3K/Akt)signaling pathway,sex hormone and insulin related index in rats with polycystic ovary syndrome(PC OS)and insulin resistance(IR).Method A dual rat model of PCOS and IR was established by in-ducing with trazole and high-fat diet.Using the method of nourishing Yin and subduing Yang combined with intervention with Diane-35 and metformin,ovarian PI3K and Akt pathway signals were measured by using immunofluorescence,and real-time fluorescence quantitative PCR was used to detect ovarian PI3K and Akt pathway signal molecule mRNA expression,the protein expressions of PI3K,Akt and insulin receptor substrate 1(IRS-1),the levels of insulin receptor(INSR),sex hormones,insulin like growth factor-1(IGF-1)and insulin sensitivity index(ISI)and the changes in ovarian tissue morphology were observed.Result The method of nourishing Yin and subduing Yang could significantly increase the relative expressions of ovarian PI3K and Akt mRNA(P<0.05),and could significantly increase the expressions of ovarian PI3K and Akt protein,up-regulate the levels of IRS-1 and INSR,reduced the levels of serum testosteron(T)and luteinizing hormone(LH)and regulate the level of ISI,and increase the number of granulosa cell layer and corpus luteum.Conclusion The method of nourishing Yin and subduing Yang can increase the signal expression levels of PI3K/Akt pathway,improve IR and follicular granulosa cell function,regulate the ratio of LH/follicle-stimulating hormone(FSH)and control the abnormal increase of LH,and regulate PCOS and IR.
